为了寻找有效预防和治疗猪繁殖与呼吸综合征(PRRS)的中草药,以 Marc 145细胞为模型,采用细胞病变(CPE)抑制试验和MTT法测定细胞活力,评价三仁汤复方中药制剂的活性成分在体外抗猪繁殖与呼吸综合征病毒(PRRSV)的作用,并通过改变药物作用方式,初步探讨复方中药制剂的抗病毒机制。结果显示,三仁汤复方中药制剂对 Marc 145细胞的最大安全质量浓度为125 g/L,在体外对PRRSV直接灭活的最小质量浓度为15.625 g/L,抑制作用的最小质量浓度为31.25 g/L,吸附阻断作用的最小质量浓度为125 g/L,其对PRRSV的灭活作用明显优于抑制作用和吸附阻断作用,吸附阻断作用最弱。表明,在安全质量浓度范围内,三仁汤中药复方制剂在体外有显著的抗PRRSV作用,可以作为预防及治疗PRRS的药物。

In order to search for Chinese medicinal herb that could be effective in preventing and treating PRRS,this research used Marc-145 cells as testing models,through cytopathic effect inhibition assay (CPE ) and cell vitality determined by MTT method to evaluate the active ingredient of three kernel soup compound preparation that showed inhibitory effect on cells infected by PRRSV in vitro.Additionally,in order to discuss preliminarily the antiviral mechanism of Chinese medicinal herbal,the measures to change the dosing way were took.The results showed that the maximum non-toxic concentration of three kernel soup compound preparation effected on Marc-145 cells was 125 g/L,the minimum concentration that it could inactivate PRRSV directly in vitro was 1 5.6 2 5 g/L,the minimal inhibitory concentration was 31.25 g/L and the minimal blocking concentration was 125 g/L.Therefore,three kernel soup compound preparation inactivated PRRSV significantly better than inhibitory and blocking effect, the weakest effect was blocking.It demonstrated that three kernel soup compound preparation had significant antiviral effect in the safety concentration range invitro,and it could be used as a drug in prevention and treatment of PRRS.
