以活菌数为检测指标,通过单因子和正交试验对淡紫拟青霉 E16液体发酵工艺进行优化。结果表明,淡紫拟青霉E16发酵最佳液体培养基配方为:白砂糖1.5%(质量分数,下同),大豆粉1%,磷酸二氢钾0.1%,硫酸锌0.01%;最佳发酵条件为:起始 pH 值6,500 mL 三角瓶装液量30%,接菌量5%,培养温度30℃,摇床转速150 r/min,培养时间4 d。在最佳发酵培养基和发酵条件下,淡紫拟青霉E16产生的活菌数为9.80×109 cfu/mL,较优化前提高近1个数量级。

이활균수위검측지표,통과단인자화정교시험대담자의청매 E16액체발효공예진행우화。결과표명,담자의청매E16발효최가액체배양기배방위:백사당1.5%(질량분수,하동),대두분1%,린산이경갑0.1%,류산자0.01%;최가발효조건위:기시 pH 치6,500 mL 삼각병장액량30%,접균량5%,배양온도30℃,요상전속150 r/min,배양시간4 d。재최가발효배양기화발효조건하,담자의청매E16산생적활균수위9.80×109 cfu/mL,교우화전제고근1개수량급。
According to the number of viable cells,the liquid fermentation technique of Paecilomyces lilacinus E16 was optimized by the single factor and orthogonal design experiments.The best liquid medium of E1 6 was determined as following:sugar 1.5%(mass fraction),soybean powder 1%,potassium dihydrogen phosphate 0.1%,zinc sulfate 0.01%.And the optimum culture conditions were determined as following:an initial pH value of 6,500 mL flasks,liquid volume of 30%,inoculum amount of 5%,culture temperature of 30 ℃,shaking speed of 150 r/min, cultivating for 4 d.Under the optimal medium and fermentation conditions,the number of viable cells of P.lilacinus E16 was 9.80×109 cfu/mL,increasing by one order of magnitude compared with before optimization.
