建立3,5-二硝基水杨酸(DNS)法测定枸杞提取液中多糖含量的方法。结果表明:测定波长为540 nm,显色剂用量为3.0 mL,沸水浴时间为5 min,稳定时间40 min以内。方法的线性、精密度和加标回收率均较为理想。该方法准确度高,且比苯酚-硫酸法更易于操作,适合工业化生产控制。

3,5-dinitrosalicylic acid (DNS) method was used to determine the content of polysaccharide in extraction solution of Lycium barbarum. The results showed that detection wavelength was 540 nm , the dosage of DNS solution was 3.0 mL, the boiling bath time was 5 min and the stable time was 40 min. The proposed method had good liner relationship, precision and average recovery rate. The method with high accuracy was easier to operate than phenol-sulfuric acid method,and it especially was suitable for determining polysaccharide in the industrial process.
