目的:探讨FAK和CD44v6在大肠癌浸润转移中的作用。方法免疫组织化学SABC法原位观察正常大肠粘膜、癌旁及癌组织中FAK和CD44v6蛋白的表达,分析两蛋白表达的相关性。结果 FAK在低分化、Duck’s C+D期及有淋巴结转移的大肠癌中的阳性表达率显著高于高中分化、Duck’s A+B期及无淋巴结转移组(P<0.05或 P<0.01).其表达与患者性别、年龄、肿瘤的大小及是否侵犯浆膜层无关(P>0.05)。CD44v6蛋白在有浆膜层侵犯、Duck’s C+D期及有淋巴结转移的大肠癌中的阳性表达率显著高于无浆膜层侵犯、Duck’s A+B期及无淋巴结转移组(P<0.05或 P<0.01)。其表达与患者的年龄、性别、肿瘤大小及分化无关(P>0.05)。FAK及CD44v6蛋白在正常组及癌前病变组中的表达均显著低于大肠癌组。Speraman相关分析示,大肠癌组织中FAK和CD44v6的表达呈显著正相关(r=0.283,p=0.002)。结论 FAK和CD44v6与大肠癌的发生、进展及浸润转移密切相关,联合检测两者在大肠癌中的表达可能有助于患者预后的判断。

목적:탐토FAK화CD44v6재대장암침윤전이중적작용。방법면역조직화학SABC법원위관찰정상대장점막、암방급암조직중FAK화CD44v6단백적표체,분석량단백표체적상관성。결과 FAK재저분화、Duck’s C+D기급유림파결전이적대장암중적양성표체솔현저고우고중분화、Duck’s A+B기급무림파결전이조(P<0.05혹 P<0.01).기표체여환자성별、년령、종류적대소급시부침범장막층무관(P>0.05)。CD44v6단백재유장막층침범、Duck’s C+D기급유림파결전이적대장암중적양성표체솔현저고우무장막층침범、Duck’s A+B기급무림파결전이조(P<0.05혹 P<0.01)。기표체여환자적년령、성별、종류대소급분화무관(P>0.05)。FAK급CD44v6단백재정상조급암전병변조중적표체균현저저우대장암조。Speraman상관분석시,대장암조직중FAK화CD44v6적표체정현저정상관(r=0.283,p=0.002)。결론 FAK화CD44v6여대장암적발생、진전급침윤전이밀절상관,연합검측량자재대장암중적표체가능유조우환자예후적판단。
Objective To explore the expression of FAK and CD44v6 proteins and its relation to invasiveness and metastasis o f colorectal carcinoma. (evaluate their clinical value.)Methods SABC immunohistochemistry was used to detect the expression of FAK and CD44v6 and analyze the relationship between them. Results The positive expression rate of FAK protein was significantly higher in colorectal carcionma with poor differation, Ducks C+D stages and lymph node metastasis than that with high or moderate differation, Ducks A+B stages and without lymph node metastasis, respectively. FAK expression had no significant correlation with patient’s age, sex, tumor size and serosa infiltration. The positive expression rate of CD44v6 was significantly higher in the group of serosa infiltration, Duck’s C+D stages and lymph node metastasis than that of no serosa infiltration, Duck’s A+B stages and without lymph node metastasis, respectively. There was no significant association with age, sex and tumor differation. There was positive correlation between the expression of FAK and CD44v6(r=0.283,p=0.002. The expression of FAK and CD44v6 was significantly lower in normal tissue and precancerous lesions than in tumor tissue. Conclusions Increased expression of FAK and CD44v6 play an important role in carcinogenesis, progression and metastasis of colorectal carcinoma. Combined detection of FAK and CD44v6 may be valuable in predicting the prognosis of patients with colorectal carcinoma.
