目的：研究过氧化物酶体增殖物激活受体γ（ PPARγ）在妊娠期糖尿病（ GDM ）脂肪及胎盘组织的表达，揭示 PPARγ参与 GDM 的病理生理机制。方法收集 GDM 及正常妊娠者的脂肪及胎盘组织，分别用 Real-time PCR 和 Western 印迹方法检测 PPARγmRNA 及蛋白水平在两组样本中的改变。结果与正常组相比， PPARγ在 GDM 脂肪组织中表达下降（ mRNA及蛋白均降低40％左右，P 值分别为0.014、0.024），而在胎盘组织中表达上调（ mRNA 增加了64％，蛋白增加至原来的2倍， P 值均为0.002）。结论 GDM 脂肪组织中 PPARγ的降低可能是 GDM 发生的重要原因，而胎盘组织中 PPARγ可能受血糖等因素的影响而产生反馈性增强；PPARγ表达的改变可能是 GDM 发病的重要机制之一。
목적：연구과양화물매체증식물격활수체γ（ PPARγ）재임신기당뇨병（ GDM ）지방급태반조직적표체，게시 PPARγ삼여 GDM 적병리생리궤제。방법수집 GDM 급정상임신자적지방급태반조직，분별용 Real-time PCR 화 Western 인적방법검측 PPARγmRNA 급단백수평재량조양본중적개변。결과여정상조상비， PPARγ재 GDM 지방조직중표체하강（ mRNA급단백균강저40％좌우，P 치분별위0.014、0.024），이재태반조직중표체상조（ mRNA 증가료64％，단백증가지원래적2배， P 치균위0.002）。결론 GDM 지방조직중 PPARγ적강저가능시 GDM 발생적중요원인，이태반조직중 PPARγ가능수혈당등인소적영향이산생반궤성증강；PPARγ표체적개변가능시 GDM 발병적중요궤제지일。
Objective Measure the expression of peroxisome proliferator-activated receptor γ( PPARγ) in the fat and planceta of gestational diabetes mellitus ( GDM ) , aim to explore the role of PPARγin pathophysiology of GDM .Methods Collect the fat and placenta tissues of GDM and normal pregnant woman .Real-time PCR and Western blot were used to measure the mRNA and protein of PPARγin the two pairs of tissues .Results Compared to the normal group , the expression of PPARγin fat was lower in GDM group , both mRNA ( P =0.0 1 4 ) and protein level ( P =0.0 2 4 ) were decreased nearly 4 0%.While in the placenta , the expression of PPARγwas higher in GDM group , the mRNA and pro-tein level ( P =0.0 0 2 ) increased 1.6-fold and 2.0-fold , respectively .Conclusion Our results sug-gested that PPARγdecreased in fat and increased in placenta of GDM cases , and PPARγmay participate in the pathophysiology of GDM .
