在液态发酵条件下,采用单因素实验确定了Aspergillus niger PZ331产异淀粉酶的最适碳源和氮源,分别为蔗糖和硝酸铵。在上述基础上利用Plackett-Burman设计对影响产异淀粉酶的因素进行评价,并筛选出硝酸铵、接种量、培养温度3个主要因素;继而利用响应面设计优化了最佳硝酸铵浓度、接种量和培养温度。最终确定了最优培养条件为:蔗糖10 g/L,硝酸铵10 g/L,磷酸氢二钾3 g/L,硫酸亚铁0.01 g/L,硫酸镁1 g/L,起始pH值4.2;接种量2%(孢子浓度为107cfu/mL),30℃培养72 h,酶活达137.3μ/mL;比基础培养基的提高了1.71倍左右。

Based on liquid fermentation , the optimum carbon and nitrogen of isoamylase for Aspergillus niger PZ331 were obtained through the single factor experiment , which were sucrose and NH 4 NO3 , respectively .Some process factors of isoamylase production were evaluated by Plackett-Burrman design, and three significant factors ( NH4 NO3 , inoculation amount and temperature ) were ob-tained.Response surface methodology was further used to optimize the three critical internal factors .The results showed that the opti-mum culture medium(g/L) of isoamylase was composed of 10 NH4NO3, 10 sucrose, 3 K2HPO4,0.01FeSO4,1 MgSO4,initial pH 4.2;Under the above conditions , Aspergillus niger PZ331 was cultured under 30℃for 72 h with inoculation amount of 2%( spore concentra-tions of 107cfu/mL), the maximum yield of isoamylase was 137.3 μ/mL, which increased by 1.71-fold.
