Pegylated recombinant human erythropoietin( PEG-EPO)%Succinimide%HPLC
目的:建立聚乙二醇重组人促红素( PEG-EPO)中丁二酰亚胺残留量测定的高效液相色谱法。方法使用高效液相色谱法,色谱柱为Shiseido Capcell PAK C18 AQ(4.6 mm ×250 mm,5μm),流动相为0.02 mol·L-1 KH2PO4(pH 6.5),检测波长为210 nm,流速为0.7 mL·min-1。结果线性范围为0.08~0.8μg(r=1),6次重复进样峰面积响应值的RSD为0.61%,平均加样回收率为100.96%。结论建立的高效液相色谱法简单、精密度良好、准确度良好,可用于PEG-EPO中丁二酰亚胺残留量检测。

목적:건립취을이순중조인촉홍소( PEG-EPO)중정이선아알잔류량측정적고효액상색보법。방법사용고효액상색보법,색보주위Shiseido Capcell PAK C18 AQ(4.6 mm ×250 mm,5μm),류동상위0.02 mol·L-1 KH2PO4(pH 6.5),검측파장위210 nm,류속위0.7 mL·min-1。결과선성범위위0.08~0.8μg(r=1),6차중복진양봉면적향응치적RSD위0.61%,평균가양회수솔위100.96%。결론건립적고효액상색보법간단、정밀도량호、준학도량호,가용우PEG-EPO중정이선아알잔류량검측。
Objective To establish a method for the determination of residual succinimide in PEG - EPO by HPLC. Methods HPLC method was used and the separation was performed on a column packed with Shiseido Capcell PAK C18 AQ(4. 6 mm × 250 mm,5 μm). The mobile phase was 0. 02 mol·L-1 KH2 PO4( pH 6. 5). The detective wave-length was set at 210 nm. The flow rate was 0. 7 mL·min-1 . Results The linear range was good in 0. 08~0. 8 μg( r=1 ). The RSD of peak area of six repeated sampling was 0. 61%. The average recovery was 100. 96%. Conclusion The es-tablished method was simple,high sensitive and accurate with a high reproducibility and can be used for the determination of residual succinimide in PEG-EPO.
