目的::确定芍药苷对人黑素细胞生物活性及迁移的影响。方法:体外培养人表皮黑素细胞,分别使用MTT法、L-DOPA法、NaOH法、Transwell小室法检测不同浓度的芍药苷对黑素细胞的增殖活性、酪氨酸酶活性、黑素合成以及黑素细胞迁移的影响。结果:芍药苷能显著促进黑素细胞增殖和上调酪氨酸酶活性,且10μg/mL是产生这两方面作用的最佳浓度;芍药苷也能促进黑素的合成,50μg/mL时作用最显著。芍药苷在20μg/mL时显著促进黑素细胞迁移。各组与空白对照组比较差异均有显著性( P<0.05)。结论:芍药苷单体可能是白芍中促进黑素细胞迁移的主要活性成分。

목적::학정작약감대인흑소세포생물활성급천이적영향。방법:체외배양인표피흑소세포,분별사용MTT법、L-DOPA법、NaOH법、Transwell소실법검측불동농도적작약감대흑소세포적증식활성、락안산매활성、흑소합성이급흑소세포천이적영향。결과:작약감능현저촉진흑소세포증식화상조락안산매활성,차10μg/mL시산생저량방면작용적최가농도;작약감야능촉진흑소적합성,50μg/mL시작용최현저。작약감재20μg/mL시현저촉진흑소세포천이。각조여공백대조조비교차이균유현저성( P<0.05)。결론:작약감단체가능시백작중촉진흑소세포천이적주요활성성분。
Objective:To determine the effects of paeoniflorin on biological activities and migration of mel ̄anocytes. Methods:A melanocytes culture system was established and proliferation activity, tyrosinase activi ̄ty, melanogenesis and migration were detected by MTT, L-DOPA, NaOH method and transwell assay respec ̄tively. Results:The paeoniflorin improved the melanocytes proliferation and tyrosinase activity, in which the effects were the most obvious at the concentration of 10 μg/mL. The paeoniflorin promoted the synthesis of melanin, which was the most obvious at the concentration of 50μg/mL. Paeoniflorin promoted the migration of melanocytes obviously at the concentration of 20μg/mL. There were significant differences between the paeon ̄iflorin groups and the control groups ( P<0.05) . Conclusion:The paeoniflorin is the principal active compo ̄nent of radix paeoniae alba that promoting the migration of melanocytes.
