目的:探讨亚低温干预下新生大鼠缺氧缺血性脑损伤(HIBD)时环氧化酶2(COX 2)基因表达变化及意义。方法120只7 d龄 SD大鼠,采用 Rice法建立新生大鼠缺氧缺血脑损伤模型,随机分为假手术组、常温组、亚低温组,每组又根据处死时间的不同分为1 h、6 h、12 h、24 h、48 h五个亚组,用实时荧光定量聚合酶链反应检测新生大鼠COX 2mRNA表达。结果假手术组COX 2mRNA表达甚微,且各组之间无明显变化;常温组 COX 2mRNA表达6 h开始上调且逐渐增加,24 h含量达高峰,24 h~48 h维持在高峰(P<0.05);亚低温组各组 COX 2mRNA表达与常温组各时间点比较均降低(P<0.05)。结论 COX 2在新生大鼠 HIBD形成中发挥一定的作用,亚低温干预能降低 HIBD后不同时间脑组织 COX 2的表达活性,提示在恰当的时间窗内进行亚低温干预治疗对脑保护是有效的。
目的:探討亞低溫榦預下新生大鼠缺氧缺血性腦損傷(HIBD)時環氧化酶2(COX 2)基因錶達變化及意義。方法120隻7 d齡 SD大鼠,採用 Rice法建立新生大鼠缺氧缺血腦損傷模型,隨機分為假手術組、常溫組、亞低溫組,每組又根據處死時間的不同分為1 h、6 h、12 h、24 h、48 h五箇亞組,用實時熒光定量聚閤酶鏈反應檢測新生大鼠COX 2mRNA錶達。結果假手術組COX 2mRNA錶達甚微,且各組之間無明顯變化;常溫組 COX 2mRNA錶達6 h開始上調且逐漸增加,24 h含量達高峰,24 h~48 h維持在高峰(P<0.05);亞低溫組各組 COX 2mRNA錶達與常溫組各時間點比較均降低(P<0.05)。結論 COX 2在新生大鼠 HIBD形成中髮揮一定的作用,亞低溫榦預能降低 HIBD後不同時間腦組織 COX 2的錶達活性,提示在恰噹的時間窗內進行亞低溫榦預治療對腦保護是有效的。
목적:탐토아저온간예하신생대서결양결혈성뇌손상(HIBD)시배양화매2(COX 2)기인표체변화급의의。방법120지7 d령 SD대서,채용 Rice법건립신생대서결양결혈뇌손상모형,수궤분위가수술조、상온조、아저온조,매조우근거처사시간적불동분위1 h、6 h、12 h、24 h、48 h오개아조,용실시형광정량취합매련반응검측신생대서COX 2mRNA표체。결과가수술조COX 2mRNA표체심미,차각조지간무명현변화;상온조 COX 2mRNA표체6 h개시상조차축점증가,24 h함량체고봉,24 h~48 h유지재고봉(P<0.05);아저온조각조 COX 2mRNA표체여상온조각시간점비교균강저(P<0.05)。결론 COX 2재신생대서 HIBD형성중발휘일정적작용,아저온간예능강저 HIBD후불동시간뇌조직 COX 2적표체활성,제시재흡당적시간창내진행아저온간예치료대뇌보호시유효적。
Objective To observe the changes of cycloxygenase 2(COX 2)gene expression in brain tissue after hypoxia is-chemia brain damage (HIBD)and receiving mild hypothermia intervention.Methods One hundred and twenty seven day old neo-natal Sprague Dawley(SD)rats that adopted Rice method to establish the model of HIBD and were randomly divided into three groups:Sham operated group (n= 40),normal temperature group (n= 40)and mild hypothermia (n= 40)group.The experimen-tal groups were further divided into five sub groups (n= 8 in each group)based on different time points after HIBD (1 h,6 h,12 h, 24 h,48 h)respectively.Fluorescence quantitative PCR (PT PCR)technique was used to determine the expression changes of COX 2 mRNA in brain tissues.Results COX 2 mRNA expressed had no significant change in sham operated group.The expres-sion of normal temperature group increased at 6 h after HIBD,24 h to the peak level,24 h to 48 h gradual y increased(P<0.05).The COX 2 mRNA expression in mild hypothermia group was significantly lower than that in HIBD group at each time points (P<0.05). Conclusion COX 2 participated in HIBD of neonatal rats.Mild hypothermia intervention could reduce the expression of COX 2 mRNA in brain tissue after HIBD.Therefore,mild hypothermia intervention at the appropriate time window had some cerebral protec-tion effects on the brain tissues of neonatal rats after HIBD.
