目的:观察纳米级甲壳胺长期灌胃给药后对大鼠可能产生的毒性反应,为临床应用提供毒理学依据。方法实验大鼠随机分为对照组和甲壳胺低、中、高剂量组;给药组分别灌胃给予纳米级甲壳胺0.5,2.5,5.0 g/(kg·d)的剂量(约为临床给药剂量的5,25,50倍),每天早、晚各给药1次,连续给药12周,停药2周。期间观察动物行为、外观及大小便。测量受试大鼠的体重,测定血液学及血液生化学指标,计算重要器官脏器系数,并进行其组织病理学检查。结果与对照组相比,给药各组大鼠给药期及停药恢复期的一般状况良好,饮食情况、体重增长、血常规、血液生化指标、脏器系数、解剖学和主要脏器的组织形态学检查均正常,未见病变。结论纳米级甲壳胺长期口服用药对大鼠无明显的毒副作用,使用安全。

목적:관찰납미급갑각알장기관위급약후대대서가능산생적독성반응,위림상응용제공독이학의거。방법실험대서수궤분위대조조화갑각알저、중、고제량조;급약조분별관위급여납미급갑각알0.5,2.5,5.0 g/(kg·d)적제량(약위림상급약제량적5,25,50배),매천조、만각급약1차,련속급약12주,정약2주。기간관찰동물행위、외관급대소편。측량수시대서적체중,측정혈액학급혈액생화학지표,계산중요기관장기계수,병진행기조직병이학검사。결과여대조조상비,급약각조대서급약기급정약회복기적일반상황량호,음식정황、체중증장、혈상규、혈액생화지표、장기계수、해부학화주요장기적조직형태학검사균정상,미견병변。결론납미급갑각알장기구복용약대대서무명현적독부작용,사용안전。
Objective To observe the possible generated toxicity reactions of chitosan nanoparticles after long term intragastric administra-tion to provide the toxicological basis for clinical application. Methods The experimental rats were randomly divided into the control groups, and low, middle and high dose chitosan groups. The chitosan groups was given chitosan with a dose of 0. 5, 2. 5, 5. 0 g/( kg·d ) re-spectively ( equaling to 5, 25, 50 times of clinical giving dose ) by intragastric administration, once at morning and night every day and for successive 12 weeks. Then the medication was withdrawn for 2 weeks. During the administration period, rats ' behavior, appearance, urine and stools were observed, the body weight was measured once every week. The hematology and biochemistry indexes were measured. Then the rats were killed, and the organ coefficients of important organs were calculated and the main organs were examined by the pathological method. Results Compared with the control group, the rats in various medication groups had better general condition during the medication period and the recovery period after medication withdrawal, the diet situation, body weight gain, blood routine, blood biochemical indicators, organ coefficients, anatomy and histomorphological examination of main organs all were normal without disease lesions. Conclusion Long-term administration of chitosan nanoparticles has no apparent toxicity and no obvious side effects on rats and its use is safe.
