目的:探讨肉苁蓉总苷(GCs)对SAM P8小鼠空间学习记忆的影响及其作用机制。方法:GCs组动物灌胃给药,100 mg/d;氟他胺(F)组动物皮下注射氟他胺0.5 mg/d;(F+GCs)组动物联合给予上述等剂量药物;Vehicle组动物给予等体积的生理盐水灌胃;以上4组动物连续给药30 d。空白对照(Control)组不给予任何干预。测定SAM P8小鼠学习记忆能力,海马CA1区正常锥体细胞数;测定脑组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性及谷胱甘肽过氧化物酶(GSH -Px)活性。结果:GCs可提高SAMP8小鼠空间学习记忆能力,可降低脑组织MDA含量,提高SOD、GSH -Px活性,提高海马锥体细胞的存活率。结论:GCs对脑损伤的保护机制可能与其增强自由基清除酶活性,减轻脂质过氧化反应,进而提高海马锥体细胞的存活率有关。

목적:탐토육종용총감(GCs)대SAM P8소서공간학습기억적영향급기작용궤제。방법:GCs조동물관위급약,100 mg/d;불타알(F)조동물피하주사불타알0.5 mg/d;(F+GCs)조동물연합급여상술등제량약물;Vehicle조동물급여등체적적생리염수관위;이상4조동물련속급약30 d。공백대조(Control)조불급여임하간예。측정SAM P8소서학습기억능력,해마CA1구정상추체세포수;측정뇌조직병이철(MDA)함량、초양화물기화매(SOD)활성급곡광감태과양화물매(GSH -Px)활성。결과:GCs가제고SAMP8소서공간학습기억능력,가강저뇌조직MDA함량,제고SOD、GSH -Px활성,제고해마추체세포적존활솔。결론:GCs대뇌손상적보호궤제가능여기증강자유기청제매활성,감경지질과양화반응,진이제고해마추체세포적존활솔유관。
Objective :To observe the effect of cistanche glycosides (GCs) on spatial learning and memory in SAMP8 mice and its mechanism .Methods :GCs group animals administered orally ,100 mg per day ;flutamide (F) group animals were injected subcutaneously F 0 .5 mg per day ,F+GCs group animals were given the same doses of GCs and F ,Vehicle group animals were given an equal volume of normal saline ,four groups above animals were administered 30 days consecu_tively ;Control group animals were not given any intervention .The spatial learning and memory of mice were observed by the Morris water maze task ;the content of MDA ,the activities of SOD and GSH -Px in mice brain were detected by kits ;the number of intact pyramidal cells of hippocampal CA1 region was detected .Results :GCs was shown to improve spatial learning and memory deficiency in SAMP8 mice;to reduce content of MDA ;to increase activities of SOD ,GSH -Px in mice brain ;to increase survival rate of intact pyramidal cells of hippocampal CA 1 region in SAM P8 mice .Conclu_sion:The protective effects of GCs on spatial learning and memory in SAMP8 mice may be involved in scavenging free radical ,antioxidation and increasing survival rate of intact pyramidal cells of hippocampal CA 1 region in SAM P8 mice .
