目的:通过动态光散射分析血红蛋白A2现象的分子机理。方法:首先,将正常成人新鲜抗凝全血制备成红细胞悬液和溶血液,利用淀粉-琼脂糖混合凝胶电泳分别分离得到红细胞和溶血液的 HbA和 HbA2,从凝胶中将这些区带分别切出,通过反复冻融,分离出区带内所含蛋白。用动态光散射方法对红细胞和溶血液的 HbA和 HbA2的粒子直径(粒径)进行分析。结果:红细胞HbA的粒径与溶血液 HbA的粒径基本相同,含量最多的粒径约为7.5 nm左右;HbA2的粒径大于HbA ,而且红细胞 HbA2的粒径(28.21 nm)明显大于溶血液 HbA2的粒径(13.54 nm)。结论:红细胞中的HbA2并不是单独存在的,而是与某些成分结合形成复合体。

목적:통과동태광산사분석혈홍단백A2현상적분자궤리。방법:수선,장정상성인신선항응전혈제비성홍세포현액화용혈액,이용정분-경지당혼합응효전영분별분리득도홍세포화용혈액적 HbA화 HbA2,종응효중장저사구대분별절출,통과반복동융,분리출구대내소함단백。용동태광산사방법대홍세포화용혈액적 HbA화 HbA2적입자직경(립경)진행분석。결과:홍세포HbA적립경여용혈액 HbA적립경기본상동,함량최다적립경약위7.5 nm좌우;HbA2적립경대우HbA ,이차홍세포 HbA2적립경(28.21 nm)명현대우용혈액 HbA2적립경(13.54 nm)。결론:홍세포중적HbA2병불시단독존재적,이시여모사성분결합형성복합체。
Objectives To analyze the molecular mechanism of hemoglobin A 2 (HbA2 ) phenomenon by dynamic light scattering (DLS) .Methods First ,prepare RBC suspension and hemolysate from fresh normal aldult whole blood .Then erythrocyte suspension and hemolysate was separated by starch -agarose mixed gel electrophoresis ,Finaly , HbA and HbA2 were acquired from the gel by freeze thawing method ,and then the size of these Hb was analyzed by DLS .Results The size of RBC HbA was the same with that of hemolysate's ,and the diameter of most of the HbA was a_bout 7 .5 nm .The size of HbA2 is larger than that of HbA ,and the size of RBC HbA2 (28 .21 nm) was much larger than that of hemolysate HbA2 (13 .54 nm) .Conclusions HbA2 is not existed alone in RBC ,but in combination with the other components .
