目的:探讨三氧化二砷(As2 O3)抑制大肠癌 SW620细胞增殖和转移的机制。方法将不同浓度的 As2 O3(0,1,2,4,8μmol∕ L)作用于人的大肠癌 SW620细胞,用逆转录_聚合酶链反应( RT_PCR)检测各组大肠癌细胞NOTCH1mRNA 表达,免疫细胞化学( SP 法)检测各组 CD133,KI67,血管内皮生长因子_C( VEGF_C)蛋白的表达。结果随着 As2 O3浓度的增加,NOTCH1mRNA、KI67、VEGF_C、CD133的表达逐渐减少,CD133与 Notch1呈正相关,差异均有统计学意义(p <0?05)。结论 As2 O3具有抑制大肠癌 SW620细胞株细胞增殖和转移的作用,其机制可能与下调Notch1,以及对 CD133、KI67、VEGF_C 的抑制有关。

목적:탐토삼양화이신(As2 O3)억제대장암 SW620세포증식화전이적궤제。방법장불동농도적 As2 O3(0,1,2,4,8μmol∕ L)작용우인적대장암 SW620세포,용역전록_취합매련반응( RT_PCR)검측각조대장암세포NOTCH1mRNA 표체,면역세포화학( SP 법)검측각조 CD133,KI67,혈관내피생장인자_C( VEGF_C)단백적표체。결과수착 As2 O3농도적증가,NOTCH1mRNA、KI67、VEGF_C、CD133적표체축점감소,CD133여 Notch1정정상관,차이균유통계학의의(p <0?05)。결론 As2 O3구유억제대장암 SW620세포주세포증식화전이적작용,기궤제가능여하조Notch1,이급대 CD133、KI67、VEGF_C 적억제유관。
Objective To investigate the mechanism of proliferation and metastasis of colorectal cancer SW620 cells inhibited by arsenic trioxide(As2 O3 )? Methods Human colorectal cancer SW620 cell were treated with different concentrations of As2 O3 (0,1,2,4,8 μmol∕ L)? Reverse transcription_polymerase chain reaction(RT_PCR)were used to detect the mRAN expression of signaling pathway NOTCH1 in each group? Immunocytochemistry was used to detect the expression of CD133,KI67,VEGF_C by different concentrations of As2 O3? Results For the concentration of As2 O3 decrease,NOTCH1 mRNA,KI67,VEGF_C,CD133 expression was gradually decreased in each group? Correlation analysis showed that a positive correlation of CD133 with NOTCH1,and the difference was statistically significant(p < 0? 05)? Conclusion Arsenic trioxide can inhibite the proliferation and metastasis of colorectal cancer SW620 cells? Its mechanisms might be probably related to the down_regulation of Notch1 ex_pression in this process,and inhibite the expression of CD133,KI67,VEGF_C.
