Gastrodia elata%β-sitosterol%n-butyl alcohol (n-butanol)%HPLC
以正丁醇为溶剂,超声提取天麻中β-谷甾醇;采用高效液相色谱(HPLC)法及 MD-ODS C18色谱柱(250 mm×4.6 mm,5μm),在流动相为100%甲醇、检测波长为210 nm、流速为0.7 mL/min、柱温35℃、进样量为20μL 时,测定不同产地天麻中β-谷甾醇含量。结果表明,β-谷甾醇在0.184~0.92μg/mL (r =0.9998)线性关系良好;平均加样回收率为98.26%,RSD 1.41%。22个不同产地天麻品种中β-谷甾醇含量最高可达到1.1248 mg/g。该方法简便快速,准确可靠,可为天麻药材质量标准提供新依据。

Mixed with n-butanol solvent,the β-sitosterol chemical composition in Gastrodia elata BI was extracted by ultrasonic.The β-sitosterol in Gastrodia elata BI from different originwas determined by HPLC column in the context of MD-ODS C1 8 column (250 mm × 4.6 mm,5 μm),mobile phase with 100% methanol,detection wavelength with 210 nm,flow rate with 0.7 mL/min,column temperature with 35℃,injection volume with 20 μL.The results showed that:when the β-sitosterol was in the 0.184 ~0.92 μg/mL (r=0.999 8),its linear relationship was good,the average recovery was 98.26 %,RSD was 1.41 %. Theβ-sitosterol in Gastrodia elata BI from 22 different origin was detected,and the maximum of theβ-sitosterol content was 1.124 8 mg/g.The method was simple,rapid,accurate and reliable,which could provide a new basis for Gastrodia elata BI medicinal quality standards.
