目的:探讨氟伐他汀(fluvastatin, Flu)对小牛血清(CS)干预的高血压大鼠胸主动脉血管平滑肌细胞增殖及血管重构的影响并初步探讨其可能的机制。方法 CS﹑ Flu 和甲羟戊酸(mevalonic acid, Me-va)处理血管平滑肌细胞4 d,将细胞分为 SHR 组﹑ SHR +20% CS 组﹑ SHR +20% CS + Flu 组和 SHR +20% CS + Flu + Meva 组,采用 MTT 检测细胞的增殖能力, Transwell 检测细胞的迁移能力,体外管样实验﹑天狼星红染色以及免疫荧光的方法分别检测细胞的管样结构的生成能力﹑胶原纤维以及 F-actin 的表达, Western 印迹检测 PMCA﹑ SERCA 和 LTCC 的表达。结果 Flu 能够抑制 CS 促进的 VSMCs 增殖效应﹑迁移能力,管样结构生成,胶原蛋白表达,以及骨架蛋白的聚合,并下调 VSMCs 中 PMCA 的表达,上调 SERCA和 LTCC 的表达,而 Meva 则对 Flu 的作用进行逆转。结论氟伐他汀对 CS 诱导的 VSMCs 增殖及血管重构产生抑制,该过程可能是通过调节钙泵的表达实现的,而 Meva 代谢途径可能在其中发挥了重要的作用。

목적:탐토불벌타정(fluvastatin, Flu)대소우혈청(CS)간예적고혈압대서흉주동맥혈관평활기세포증식급혈관중구적영향병초보탐토기가능적궤제。방법 CS﹑ Flu 화갑간무산(mevalonic acid, Me-va)처리혈관평활기세포4 d,장세포분위 SHR 조﹑ SHR +20% CS 조﹑ SHR +20% CS + Flu 조화 SHR +20% CS + Flu + Meva 조,채용 MTT 검측세포적증식능력, Transwell 검측세포적천이능력,체외관양실험﹑천랑성홍염색이급면역형광적방법분별검측세포적관양결구적생성능력﹑효원섬유이급 F-actin 적표체, Western 인적검측 PMCA﹑ SERCA 화 LTCC 적표체。결과 Flu 능구억제 CS 촉진적 VSMCs 증식효응﹑천이능력,관양결구생성,효원단백표체,이급골가단백적취합,병하조 VSMCs 중 PMCA 적표체,상조 SERCA화 LTCC 적표체,이 Meva 칙대 Flu 적작용진행역전。결론불벌타정대 CS 유도적 VSMCs 증식급혈관중구산생억제,해과정가능시통과조절개빙적표체실현적,이 Meva 대사도경가능재기중발휘료중요적작용。
Objective To investigate the effect of fluvastatin (Flu) induced by CS on the pro-liferation of vascular smooth muscle cells (VSMCs) and the rearrangement of vascular and studies the possible mechanism. Methods We divided VSMCs into four groups: SHR group, SHR +20% CS group, SHR + 20% CS + Flu group; and SHR + 20% CS + Flu + Meva group. The prolifera-tion of VSMCs was detected by MTT assay, and the migratory ability changes were analyzed in tran-swell. We analyzed tube formation ability in Matrigel, and examined collagen used sirius red saturat-ed picric acid staining, and detected the expression of F-actin by immunofluorescence. Expressions of PMCA, SERCA and LTCC were analyzed by Western blot. Results Fluvastatin can inhibit the proliferative rate, migratory ability, the formation of tube, the expression of collagen and F-actin which was promoted by CS. The Western blot results showed that fluvastatin could down-regulate the expression of PMCA and up-regulated the expression of SERCA and LTCC. Conclusion Fluvasta-tin can suppress the proliferation and rearrangement of vascular effect of VSMCs which were induced by CS, it is may be achieved through the expression of Ca2 + ATPase, and the mevalonate pathway may be involved in this process.
