目的:研究姜黄素对小鼠胚胎着床,及其妊娠期子宫内膜细胞凋亡的分子调控机制。方法将见阴栓昆明白小鼠随机分为5组,连续隔日腹腔注射姜黄素,第7天(D7)回收组织进行 RT-PCR 和 West-ern 印迹检测。结果与姜黄素低剂量组相比,姜黄素中剂量组平均妊娠胎仔数显著升高(P ﹤0.05),而在高剂量组则显著下降(P ﹤0.05),且妊娠率随之下降。基因检测结果表明,凋亡因子 PTEN﹑ Caspase-3的相对 mRNA 水平随着姜黄素剂量的升高显著升高(低剂量组,中剂量组 P ﹤0.05;高剂量组 P ﹤0.01)。而核转录因子 NF-ΚB 的表达则显著降低(低剂量组,中剂量组 P ﹤0.05;高剂量组 P ﹤0.01)。且蛋白水平的检测与基因水平的检测结果相一致。结论姜黄素在小鼠胚胎着床及妊娠期子宫内膜细胞凋亡中起重要的调控作用,这对于寻找新的生殖调控手段具有重要意义。

목적:연구강황소대소서배태착상,급기임신기자궁내막세포조망적분자조공궤제。방법장견음전곤명백소서수궤분위5조,련속격일복강주사강황소,제7천(D7)회수조직진행 RT-PCR 화 West-ern 인적검측。결과여강황소저제량조상비,강황소중제량조평균임신태자수현저승고(P ﹤0.05),이재고제량조칙현저하강(P ﹤0.05),차임신솔수지하강。기인검측결과표명,조망인자 PTEN﹑ Caspase-3적상대 mRNA 수평수착강황소제량적승고현저승고(저제량조,중제량조 P ﹤0.05;고제량조 P ﹤0.01)。이핵전록인자 NF-ΚB 적표체칙현저강저(저제량조,중제량조 P ﹤0.05;고제량조 P ﹤0.01)。차단백수평적검측여기인수평적검측결과상일치。결론강황소재소서배태착상급임신기자궁내막세포조망중기중요적조공작용,저대우심조신적생식조공수단구유중요의의。
Objective To examine the effects of curcumin on the implantation of mouse em-bryos and the molecular regulation of curcumin on the apoptosis of gestational endometri-um. Methods The Kunming White mice that had the vaginal plug observed were randomly divided into five groups. Curcumin was administrated continuously by peritoneal injection, once every other day, and the gestational endometrium was removed for the detection by RT-PCR and Western blot-ting on the seventh day (D7) . Results The average gestational litter size was increased signifi-cantly (P ﹤ 0. 05) in the moderate-dose curcumin group when compared with the low-dose curcumin group, and it was decreased significantly in the high-dose curcumin group (P ﹤ 0. 01) and at the same time the pregnancy rate was decreased as well. Real-time PCR showed a significant concentra-tion-dependent increase in the mRNA expression of apoptosis-related factors PTEN and caspase-3 in curcumin groups (low-dose group vs. moderate-dose group, P ﹤ 0. 05; high-dose group vs. low- or moderate-dose group, P ﹤ 0. 01) . In contrast, the mRNA expression of NF-ΚB decreased signifi-cantly with the dose of curcumin increasing (low-dose group vs. moderate-dose group, P ﹤ 0. 05;high-dose group vs. low- or moderate-dose group, P ﹤ 0. 01) . Analysis of protein expression showed a consistent result with that by the real-time PCR. Conclusion Curcumin plays an important regula-tive role in the implantation of mouse embryos and the apoptosis of endometrial cells, which is of important significance in seeking a new reproductive regulation method.
