目的:通过免疫组织化学法实验观察 PARP‐1和 HMGA2在老年局部晚期三阴型乳腺癌中的表达,探讨其在乳腺癌中表达的临床特点及其与预后的相关性。方法采用S‐P免疫组织化学法检测80例三阴型老年局部晚期乳腺癌肿瘤标本和80例正常乳腺组织中PARP‐1与 HMGA2蛋白的表达情况。分析 PARP‐1和HMGA2在乳腺癌与正常乳腺组织中的表达差异;分析乳腺癌组织中PARP‐1与 HMGA2表达的相关性及其分别与原发肿瘤直径、腋窝淋巴结转移及病理组织学分级的相关性。并且通过Kaplan‐M eier生存曲线法比较两者阳性组和阴性组患者的预后情况。结果在乳腺癌中PARP‐1阳性表达率为84%显著高于正常乳腺组织的21%;HMGA2阳性表达率为61%高于正常乳腺组织的26.3%,PARP‐1与 HMGA2在老年局部晚期三阴型乳腺癌中的阳性表达与腋窝淋巴结转移情况及5年无瘤生存时间具有相关性(P<0.05)。PARP‐1与 HMGA2在各分子亚型乳腺癌组织中的表达具有相关性(P<0.05)。结论 PARP‐1与 HMGA2在老年局部晚期乳腺癌的浸润、转移中存在相互作用,是反映乳腺癌进展的重要生物学指标。PARP‐1与HMGA2高表达的患者易发生淋巴结转移,并且无瘤生存时间缩短,预后较差。

Objective Through the immunohistochemical method experiment ,this paper aims to observe the expressions of PARP‐1 and HMGA2 in senile patients with triple‐negative locally advanced breast cancer and to discuss their clinical significance .Methods With the S‐P immunohistochemical method ,it detected the PARP‐1 and HMGA2 protein expressions in both 80 cases of breast cancer postoperative tumor specimens and another 80 cases of normal breast tissue with a distance of at least 5cm from the tumor .We analyzed the expression difference of PARP‐1 and HMGA2 in breast cancer and normal breast tissue .Spearman rank test was used to analyze the correlation of their expressions ,and the correlation with primary tumor diameter ,auxiliary lymph node metastasis and pathological classification ,respectively .We also made a comparison of the prognosis between the positive and negative patient groups through the Kaplan‐Meier survival curve method .Results It showed that the positive ex‐pression rate of PARP‐1 in breast cancer (84% ) was significantly higher than that in paracancerous normal tissues (21% ) ,while the positive expression rate of PARP‐1 in breast cancer (61% ) was higher than that in paracancer‐ous normal tissues (26% ) .Two of them had remarkable relation to the lymph node metastasis and disease‐free survival time of 5 years(P<0.05) .Besides ,the expression of PARP‐1 and that of HMGA2 were obviously relat‐ed(P<0.05) .Conclusion The expressions of PARP‐1 and HMGA2 play cooperative roles in the infiltration and metastasis of breast cancer ,which also serve as important biological parameters for judging the breast cancer pro‐gression .Patients with up‐regulated expression of PARP‐1 and HMGA2 are prone to have lymph node metastasis and poor prognosis after operation .
