目的:观察托吡酯( TPM)对妊娠小鼠的胚胎毒性作用,探讨叶酸( FA)对其干预的效应。方法小鼠随机分为5组,每组雌雄比例2∶1,分别为:空白对照组、TPM低剂量组、TPM高剂量组、TPM低剂量+FA组、TPM高剂量+FA组,每组20只。分组后第2天给雌性鼠药物,直至妊娠结束。以阴栓出现日为妊娠零天,观察小鼠生殖能力、生长发育、畸形等指标。结果 TPM高剂量组对小鼠的生殖能力有明显毒性,补充叶酸并不能有效拮抗这种毒性作用。 TPM低剂量对妊娠期小鼠的生殖毒性作用较小。 TPM对胎鼠的生长发育有明显的影响,与剂量高低无明显关联,补充叶酸能有效拮抗低剂量TPM的毒性作用,而对高剂量TPM无明显的拮抗作用。高剂量的TPM致畸作用明显,补充叶酸能有效的拮抗致畸作用。结论 TPM对小鼠胚胎有毒性作用,补充叶酸可有效的干预。

목적:관찰탁필지( TPM)대임신소서적배태독성작용,탐토협산( FA)대기간예적효응。방법소서수궤분위5조,매조자웅비례2∶1,분별위:공백대조조、TPM저제량조、TPM고제량조、TPM저제량+FA조、TPM고제량+FA조,매조20지。분조후제2천급자성서약물,직지임신결속。이음전출현일위임신령천,관찰소서생식능력、생장발육、기형등지표。결과 TPM고제량조대소서적생식능력유명현독성,보충협산병불능유효길항저충독성작용。 TPM저제량대임신기소서적생식독성작용교소。 TPM대태서적생장발육유명현적영향,여제량고저무명현관련,보충협산능유효길항저제량TPM적독성작용,이대고제량TPM무명현적길항작용。고제량적TPM치기작용명현,보충협산능유효적길항치기작용。결론 TPM대소서배태유독성작용,보충협산가유효적간예。
Objective To observe the embryonic toxicity induced by topiramate (TPM) in pregnant mice,and to explore the intervention effect of folic acid (FA).Methods The mice were randomly divided into 5 groups,with the ratio of females/males being 2∶1,blank control group,TPM low-dose group,TPM high-dose group,TPM low-dose +FA group,TPM high-dose +FA group,with 20 mice in each group.The female mice were given drugs at 2 days after grouping until the end of pregnancy .The indexes including reproductive ability , growth and development , deformity of mice were observed for the five groups.Results As compared with that in blank control group , the live fetus rate was significantly decreased , however absorption fetus rate and dead fetus rate were significantly increased in TPM high-dose group and TPM high-dose +FA group ( P <0.01).As compared with those in blank control group ,the body weight,body length and tail length of fetal mice were significantly decreased in TPM low-dose group,TPM high-dose group,TPM high-dose +FA group ( P <0.01).As compared with that in blank control group,the abnormality rate was obviously increased in TPM high-dose group ( P <0.01),moreover, which in TPM high-dose group was significantly increased , as compared with that in TPM high-dose +FA group ( P <0.05).Conclusion TPM has toxic effect on fetal mice ,however,supplement of folic acid can effectively interfere the effect .
