目的:探讨生物安全实验室采用气化过氧化氢( H2 O2)消毒时H2 O2的最佳浓度及剂量。方法选择生物安全实验室一处,体积约120 m3,通过气化H2 O2消毒灭菌器( VHP)将H2 O2溶液气化,采用9种不同的H2 O2注入速度及维持时间组合进行实验室消毒。消毒结束后,收集在实验室10个位置点放置的化学指示卡,观察化学指示卡颜色,收集各点放置的枯草杆菌黑色变种芽胞菌片,观察各点芽胞菌片平板的菌落生长情况。观察9种组合方法消毒后各实验点化学指示卡变化、生物指示菌片变化情况,选取消毒效果合格的下最少H2 O2剂量。按照最少剂量的参数设置再重复进行6次消毒实验,以验证消毒效果。结果 H2 O2使用剂量≥5 g/m3时,消毒实验后所有化学指示卡均由蓝色变为米黄色,消毒合格。选定最佳的H2 O2使用剂量为5 g/m3,6次消毒验证实验后,消毒效果全部合格。结论生物安全实验室采用气化H2 O2消毒时最佳浓度为35%,最佳剂量为5g/m3。

Objective To design the best dosage of vapor hydrogen peroxide in disinfection of high pathogenic micro -organism biosafety laboratory .Methods One room of 120 m3 in biosafety laboratory was selected .Hydrogen peroxide solu-tion was vaporized by hydrogen peroxide sterilizer ( VHP) .Nine types of vapor hydrogen peroxide inject speed and duration time were used to disinfect .After the disinfection , the chemical indicators and biological indicators were placed on 10 loca-tions in the room to observe the color of chemical indicators and the growth condition of all the plates inoculated with Bacil -lus subtilis var niger .After the 9 types of disinfection , the minimum vapor hydrogen peroxide was determined and then re-peated for 6 times in order to verify the disinfection effect .Results Qualified disinfection was proved that the color of the chemical indicators changed from orchid to ecru when the dosage no less than 5 g/M3 .The minimum vapor hydrogen perox-ide was determined at 5 g/m3 .After 6 times repeation of disinfection experiment at 5 g/m3 , all the disinfection results were qualified.Conclusions The best dosage of hydrogen peroxide solution with concentration of 35% was 5 g/m3 .It can reach the good efficacy in disinfection of air and surfaces in biosafety laboratory , which was suit for the disinfection in high pathogenic microorganism biosafety laboratory .
