目的:检测 mTOR/p70S6K 信号通路在冠心病患者外周血 T 细胞中的表达及活化情况,探讨其在冠状动脉粥样硬化发生发展中的作用。方法:采用 Western blot 及 RT-PCR 法检测冠心病组( AMI、UAP 及 SAP )及胸痛综合征( CPS )对照组患者外周血 T 细胞中 mTOR 及其下游分子 p70S6K 基因及蛋白表达以及 p70S6K 磷酸化水平。结果:RT-PCR 结果示 mTOR、p70S6K 基因在 AMI、UAP 组表达量均显著高于 SAP 及 CPS 组( P <0.05);Western blot 结果表明, mTOR、p70S6K 蛋白表达及 p70S6K磷酸化水平在 AMI、UAP 组均显著高于 SAP 及 CPS 组( P <0.05)。结论:冠心病尤其是 AMI 及 UAP 组患者 T 细胞中存在异常活化的 mTOR 信号, mTOR/p70S6K 信号通路在冠状动脉粥样硬化发病中发挥重要作用。
目的:檢測 mTOR/p70S6K 信號通路在冠心病患者外週血 T 細胞中的錶達及活化情況,探討其在冠狀動脈粥樣硬化髮生髮展中的作用。方法:採用 Western blot 及 RT-PCR 法檢測冠心病組( AMI、UAP 及 SAP )及胸痛綜閤徵( CPS )對照組患者外週血 T 細胞中 mTOR 及其下遊分子 p70S6K 基因及蛋白錶達以及 p70S6K 燐痠化水平。結果:RT-PCR 結果示 mTOR、p70S6K 基因在 AMI、UAP 組錶達量均顯著高于 SAP 及 CPS 組( P <0.05);Western blot 結果錶明, mTOR、p70S6K 蛋白錶達及 p70S6K燐痠化水平在 AMI、UAP 組均顯著高于 SAP 及 CPS 組( P <0.05)。結論:冠心病尤其是 AMI 及 UAP 組患者 T 細胞中存在異常活化的 mTOR 信號, mTOR/p70S6K 信號通路在冠狀動脈粥樣硬化髮病中髮揮重要作用。
목적:검측 mTOR/p70S6K 신호통로재관심병환자외주혈 T 세포중적표체급활화정황,탐토기재관상동맥죽양경화발생발전중적작용。방법:채용 Western blot 급 RT-PCR 법검측관심병조( AMI、UAP 급 SAP )급흉통종합정( CPS )대조조환자외주혈 T 세포중 mTOR 급기하유분자 p70S6K 기인급단백표체이급 p70S6K 린산화수평。결과:RT-PCR 결과시 mTOR、p70S6K 기인재 AMI、UAP 조표체량균현저고우 SAP 급 CPS 조( P <0.05);Western blot 결과표명, mTOR、p70S6K 단백표체급 p70S6K린산화수평재 AMI、UAP 조균현저고우 SAP 급 CPS 조( P <0.05)。결론:관심병우기시 AMI 급 UAP 조환자 T 세포중존재이상활화적 mTOR 신호, mTOR/p70S6K 신호통로재관상동맥죽양경화발병중발휘중요작용。
Objective To investigate whether mTOR/p70S6K signal was activated in peripheral T lymphocytes of the coronary heart disease (CHD) patients and to investigate its possible role in the pathogenesis of coronary atherosclerosis. Methods The mRNA expressions of mTOR andp70S6K were detected by RT-PCR and protein expressions of mTOR, p70S6K and phosphorylated-p70S6K (p-p70S6K) were detected by Western blot assay in T cells isolated from peripheral blood of the CHD patients (including AMI, UAP, SAP) and the CPS patients. Results Levels of mTOR and p-p70S6K were significantly enhanced in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05, respectively). The mRNA expressions of mTOR and p70S6K were significantly increased in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05). Conclusion The mTOR/p70S6K pathway is activated in the peripheral T lymphocytes of CHD patients with AMI and UAP , which may participate in the pathological process of coronary atherosclerosis.
