目的:研究耳蜗内毛细胞带状突触在发育期铁缺乏过程中的变化特点。方法用普通饲料喂饲成熟雌性大鼠1周后,将其分成对照组和铁缺乏组,并分别给予普通饲料和铁缺乏饲料,2周后交配。仔鼠出生21 d断乳,出生40 d麻醉后行ABR听阈检测;ABR检测后,仔鼠经心脏采血,检测血红蛋白、平均红细胞容积、血清铁蛋白和血清铁;然后留取耳蜗,基底膜剥离,分别用CtBP2和VGLUT3进行免疫荧光标记,Dapi染核,激光共聚焦显微镜观察。结果发育期铁缺乏致仔鼠血清铁蛋白明显降低(P<0.05),而血红蛋白、平均红细胞容积和血清铁均无明显改变(P>0.05);发育期铁缺乏并未明显改变仔鼠ABR听阈(P>0.05),但P3与P1波间期延长(P<0.05)。另外,发育期铁缺乏降低CtBP2、VGLUT3表达水平。结论发育期铁缺乏可引起仔鼠内毛细胞带状突触形态和功能改变。
目的:研究耳蝸內毛細胞帶狀突觸在髮育期鐵缺乏過程中的變化特點。方法用普通飼料餵飼成熟雌性大鼠1週後,將其分成對照組和鐵缺乏組,併分彆給予普通飼料和鐵缺乏飼料,2週後交配。仔鼠齣生21 d斷乳,齣生40 d痳醉後行ABR聽閾檢測;ABR檢測後,仔鼠經心髒採血,檢測血紅蛋白、平均紅細胞容積、血清鐵蛋白和血清鐵;然後留取耳蝸,基底膜剝離,分彆用CtBP2和VGLUT3進行免疫熒光標記,Dapi染覈,激光共聚焦顯微鏡觀察。結果髮育期鐵缺乏緻仔鼠血清鐵蛋白明顯降低(P<0.05),而血紅蛋白、平均紅細胞容積和血清鐵均無明顯改變(P>0.05);髮育期鐵缺乏併未明顯改變仔鼠ABR聽閾(P>0.05),但P3與P1波間期延長(P<0.05)。另外,髮育期鐵缺乏降低CtBP2、VGLUT3錶達水平。結論髮育期鐵缺乏可引起仔鼠內毛細胞帶狀突觸形態和功能改變。
목적:연구이와내모세포대상돌촉재발육기철결핍과정중적변화특점。방법용보통사료위사성숙자성대서1주후,장기분성대조조화철결핍조,병분별급여보통사료화철결핍사료,2주후교배。자서출생21 d단유,출생40 d마취후행ABR은역검측;ABR검측후,자서경심장채혈,검측혈홍단백、평균홍세포용적、혈청철단백화혈청철;연후류취이와,기저막박리,분별용CtBP2화VGLUT3진행면역형광표기,Dapi염핵,격광공취초현미경관찰。결과발육기철결핍치자서혈청철단백명현강저(P<0.05),이혈홍단백、평균홍세포용적화혈청철균무명현개변(P>0.05);발육기철결핍병미명현개변자서ABR은역(P>0.05),단P3여P1파간기연장(P<0.05)。령외,발육기철결핍강저CtBP2、VGLUT3표체수평。결론발육기철결핍가인기자서내모세포대상돌촉형태화공능개변。
Objective To observe the effect of iron deficiency during pregnancy and lactation on ribbon synapses of cochlear inner hair cells in rats.Methods After ordinary diet for one week ,the mature female rats were divided into the control group and the iron deficiency group ,and respectively given normal diet and iron deficient diet.After two weeks ,animals were placed for mating at the same time.Newborn rats were weaned on postnatal day 21.On postnatal day 40 ,they were anesthetized ,and their auditory brainstem response (ABR)was examined. After ABR measurements ,blood samples were collected by cardiac puncture for detection of hemoglobin (Hb) ,mean corpuscular volume (MCV) ,serum ferritin (SF)and serum iron (SI). Then , cochlea was collected ,basement membranes were stripped for immunofluorescence labeled with C‐terminal binding protein 2 (CtBP2) or vesicle glumate transporter 3 (VGLUT3) ,the nuclei were stained with Dapi in the slide ,and a confocal laser scan‐ning microscope was used for observation.Results In growing period ,iron deficiency significantly decreased SF of the pups (P<0.05) ,but did not change the Hb ,MCV and SI (P>0.05);iron deficiency did not induce ABR threshold shifts (P>0.05) , but significantly increased intervals between P3 and P1 (P<0.05).In addition ,iron deficiency decreased the expression of Ct‐BP2 and VGLUT3.Conclusion Iron deficiency in growing period could induce the changes of number and function of ribbon synapses of cochlear inner hair cells in newborn rats.
