为了得到符合转录组测序要求的软枣猕猴桃花蕾RNA ,比较了Trizol法、十二烷基苯磺酸钠(SDS )法、天根植物总RNA提取试剂盒以及改良十六烷基三甲基溴化铵(CTAB )法的提取效果。结果表明,Trizol法、SDS法和天根植物总RNA提取试剂盒均不能有效提取到软枣猕猴桃花蕾总RNA ,改良后的CTAB法提取的RNA 28S和18S条带清晰,通过RT -PCR方法扩增出软枣猕猴桃Actin基因片段。将纯化后的RNA送样至测序公司,经深度检测样品合格,可以上机进行转录测序。

Quality and purity of the RNA obtained from the Actinidia arguta flower bud was important for High -throughput sequencing .In this study ,several RNA extraction protocols were compared ,including Trizol reagent ,SDS method ,total RNA extraction kit and a modified CTAB method .The results showed that only the modified CTAB method could extract total RNA from the Actinidia arguta flower bud .28S and 18S bands were clearly by electrophoresis .RT-PCR analysis of Actin gene demonstrated that the isolated RNA samples were qualified and with the transcriptional activity .The purified RNA could meet the requirements of RNA -Seq .
