目的:建立柱前衍生HPLC法测定瓜蒌皮中精氨酸含量的方法。方法:采用邻苯二甲醛( OPA )为衍生化试剂,色谱柱为Eclipse XDB C18(4.6mm ×250mm,5μm);流动相A为甲醇-乙腈-水(45∶45∶10),流动相B为0.01mol/L磷酸氢二钠-0.01mol/L硼酸钠的缓冲溶液(pH=8.2),梯度洗脱,流速为1mL/min;检测波长为338nm。结果:该方法可以对瓜蒌皮中精氨酸含量准确定量,其浓度在12~4000mg/L范围内,线性关系良好(R2=0.9999)。方法精密度、重复性均小于2%,平均加样回收率为102.36%,RSD为1.89%(n=5)。10批不同产地瓜蒌皮中精氨酸测定结果差异较大,其中,山东(道地产区)瓜蒌皮中精氨酸含量最多。结论:该方法衍生方便、快速,产物单一,测定结果准确、可靠,可用于瓜蒌皮中精氨酸的含量测定。

Objective:To establish a pre-column derivatization method of HPLC to determine the constituent of arginine in Trichosanthes pericarpium.Methods:Orthophthalaldehyde ( OPA) was used as a derivatization reagent.Chromato-graphic condition:Eclipse XDB C18 HPLC column, (4.6mm ×250mm,5μm), mobile phase:gradient elution of metha-nol-acetonitrile-water (45∶45∶10) (A) and disodium hydrogen phosphate-sodium borate buffer (ca.0.01 mol/L, pH=8.2) (B), column temperature:30℃, detector wavelength:338nm.Results:The method was simple and accu-rate with good reproducibility and could be used for the quantitative analysis of arginine in Trichosanthes pericarpium.It had a good linear relationship in the scope of 12~4 000mg/L( R2 =0.999 9) .The RSD of the method in precision and repeatability was less than 2%.The average recovery was 102.36%, RSD was 1.89%(n=5).The contents of argi-nine in 10 batches of Trichosanthes pericarpium were different, among which the content of arginine in Shandong ( real estate areas) was the highest.Conclusion:The method is accurate, reliable and sensitive, which can be used for the quantitative analysis of arginine in Trichosanthes pericarpium.
