[目的]研究白藜芦醇(Resveratrol)对脂多糖(Lipopolysacchride,LPS)诱导急性肺损伤大鼠高迁移率族蛋白(high mobility group box 1,HMGB1)的表达的影响。[方法]雄性SD大鼠,随机分为正常组(n=10)、急性肺损伤组(n=10)、白藜芦醇干扰组(n=10)。正常组用生理盐水腹腔给药及气管滴注。急性肺损伤组用生理盐水腹腔给药,用5mg·kg-1 LPS气管滴注造模。白藜芦醇干扰组造模前4d早晚各用30 mg·kg-1的白藜芦醇腹腔给药,再用5 mg·kg-1 LPS气管滴注造模。造模后24h取肺组织病理检查判定造模是否成功,收集肺泡灌洗液(Bronchoalveolar lavage fluid,BALF),离心后,上清用ELISA检测HMGB1和白介素6(IL-6)水平。从BALF离心后的细胞中分离肺泡巨噬细胞(Alveolar macrophages,AM)培养,用RT-PCR法检测肺泡巨噬细胞HMGB1 mRNA表达水平,用细胞免疫荧光法检测HMGB1蛋白在细胞内的核浆定位。[结果]白藜芦醇能明显抑制HMGB1在BALF中的释放,抑制HMGB1在AM中的表达,并抑制其从核内转位至胞浆。[结论]白藜芦醇能从mRNA和蛋白水平抑制HMGB1表达和释放,降低急性肺损伤(Acute lung inury,ALI)的炎症反应程度,具有抗炎作用。

[목적]연구백려호순(Resveratrol)대지다당(Lipopolysacchride,LPS)유도급성폐손상대서고천이솔족단백(high mobility group box 1,HMGB1)적표체적영향。[방법]웅성SD대서,수궤분위정상조(n=10)、급성폐손상조(n=10)、백려호순간우조(n=10)。정상조용생리염수복강급약급기관적주。급성폐손상조용생리염수복강급약,용5mg·kg-1 LPS기관적주조모。백려호순간우조조모전4d조만각용30 mg·kg-1적백려호순복강급약,재용5 mg·kg-1 LPS기관적주조모。조모후24h취폐조직병리검사판정조모시부성공,수집폐포관세액(Bronchoalveolar lavage fluid,BALF),리심후,상청용ELISA검측HMGB1화백개소6(IL-6)수평。종BALF리심후적세포중분리폐포거서세포(Alveolar macrophages,AM)배양,용RT-PCR법검측폐포거서세포HMGB1 mRNA표체수평,용세포면역형광법검측HMGB1단백재세포내적핵장정위。[결과]백려호순능명현억제HMGB1재BALF중적석방,억제HMGB1재AM중적표체,병억제기종핵내전위지포장。[결론]백려호순능종mRNA화단백수평억제HMGB1표체화석방,강저급성폐손상(Acute lung inury,ALI)적염증반응정도,구유항염작용。
Objective] To investigate the effect of Resveratrol on the expression of high mobility group box1 in lipopolysaccharide-induced acute lung injury rat, and explore its molecular mechanism. [Methods] 30 male SD rats were randomly divided into the normal group(n=10), ALI model group(n=10), Resveratrol group(n=10). The normal group were treated with saline instead of resveratrol intraperitoneal y administration and with saline instead of LPS tracheal instil ation, acute lung injury group were treated with saline instead of resveratrol intraperitoneal y administration and with a dose of 5 mg· kg-1 LPS tracheal instil ation to induce the acute lung injury. Resveratrol group were treated with a dose of 30 mg·kg-1 resveratrol intraperitoneal administration for four mornings and evenings each before, then with a dose of 5 mg·kg-1 LPS tracheal instil ation to model acute lung injury. Lung histopathology was carried out to judge the success of acute lung injury model. Bronchoalveolar lavage fluid was col ected, the protein level of HMGB and IL6 were detected by ELISA in the centrifuged supernatant. The alveolar macrophages were separated and cultured from BALF after centrifugation. The mRNA of HMGB1 was measured with RT-PCR and the cel location of HMGB1 was detected with immuno-fuorescence staining. [Result] Resveratrol significantly inhibited the release of HMGB1 in the BALF and the expression in the AM, furthermore prevent its translocation from the nucleus to the cytoplasm. [Conclusion] Resveratrol can inhibit the expression and release of HMGB1 and have anti-inflammatory effect.
