为分析碳源对嗜热β-半乳糖苷酶在组成型重组毕赤酵母中的表达水平,葡萄糖、果糖、丙三醇、乳糖、蔗糖和麦芽糖分别用于配制培养基的碳源。当以单糖和甘油为碳源时,酵母的生长和重组嗜热β-半乳糖苷酶的分泌水平要优于双糖;以果糖为碳源时,重组酵母分泌嗜热β-半乳糖苷酶的水平最高,其次为葡萄糖和丙三醇,蔗糖要高于麦芽糖,乳糖最低。
為分析碳源對嗜熱β-半乳糖苷酶在組成型重組畢赤酵母中的錶達水平,葡萄糖、果糖、丙三醇、乳糖、蔗糖和麥芽糖分彆用于配製培養基的碳源。噹以單糖和甘油為碳源時,酵母的生長和重組嗜熱β-半乳糖苷酶的分泌水平要優于雙糖;以果糖為碳源時,重組酵母分泌嗜熱β-半乳糖苷酶的水平最高,其次為葡萄糖和丙三醇,蔗糖要高于麥芽糖,乳糖最低。
위분석탄원대기열β-반유당감매재조성형중조필적효모중적표체수평,포도당、과당、병삼순、유당、자당화맥아당분별용우배제배양기적탄원。당이단당화감유위탄원시,효모적생장화중조기열β-반유당감매적분비수평요우우쌍당;이과당위탄원시,중조효모분비기열β-반유당감매적수평최고,기차위포도당화병삼순,자당요고우맥아당,유당최저。
To analyze the thermostabiliuy of recombinant β - Galactosidase secretory expression level in the constitutive recombinant Pichia pastoris ,glucose ,fructose - glycerol ,lactose ,sucrose and maltose were used as carbon sources respectively . The results showed that monosaccharide and glycerol were better than disaccharide in cell growth and secretory expression of recombinant β -Galactosidase .The secretory expression level of thermostability recombinant β -Galactosidase in fructose was best than the other 5 carbon resources ,glucose second and followed by glycerol ,sucrose better than maltose ,lactose was the lowest .
