目的:研究中药丹参化学成分丹参酮ⅡA(TanshinoneⅡA,TⅡA)对小鼠腹膜纤维化模型的影响。方法50只小鼠随机分为空白对照组、模型组、TⅡA 低剂量组、TⅡA 中剂量组与TⅡA 高剂量组,每组10只动物。模型组与TⅡA 组连续腹腔注射腹透液30d,TⅡA 组同时腹腔注射不同剂量丹参酮ⅡA 磺酸钠。取腹壁组织、血清与腹腔透析液,分别检测羟脯氨酸含量、腹透1hr透析液/透析液起始葡萄糖浓度比值( D1/D0 GLU)与透析液/血清尿素浓度比值(D/P BUN),比较各组之间的差异。结果与空白对照组相比,模型组腹壁羟脯氨酸含量增加,D/P BUN值明显升高,D1/D0 GLU值降低,统计学差异显著(P<0.01),说明腹膜纤维化模型制备成功。 TⅡA 中、高剂量组羟脯氨酸含量均低于模型组,统计学差异显著(P<0.01),与空白对照组相比无统计学差异(P>0.05)。 TⅡA 中、高剂量组D1/D0 GLU值均高于模型组,统计学差异显著(P<0.01)。 TⅡA 低、中、高剂量组D/P BUN值虽比模型组有所降低,但无统计学差异(P>0.05)。由此推断丹参酮ⅡA 具有改善腹膜功能以及减少胶原沉积的作用。结论丹参酮ⅡA 为丹参预防腹膜纤维化的有效成分。

목적:연구중약단삼화학성분단삼동ⅡA(TanshinoneⅡA,TⅡA)대소서복막섬유화모형적영향。방법50지소서수궤분위공백대조조、모형조、TⅡA 저제량조、TⅡA 중제량조여TⅡA 고제량조,매조10지동물。모형조여TⅡA 조련속복강주사복투액30d,TⅡA 조동시복강주사불동제량단삼동ⅡA 광산납。취복벽조직、혈청여복강투석액,분별검측간포안산함량、복투1hr투석액/투석액기시포도당농도비치( D1/D0 GLU)여투석액/혈청뇨소농도비치(D/P BUN),비교각조지간적차이。결과여공백대조조상비,모형조복벽간포안산함량증가,D/P BUN치명현승고,D1/D0 GLU치강저,통계학차이현저(P<0.01),설명복막섬유화모형제비성공。 TⅡA 중、고제량조간포안산함량균저우모형조,통계학차이현저(P<0.01),여공백대조조상비무통계학차이(P>0.05)。 TⅡA 중、고제량조D1/D0 GLU치균고우모형조,통계학차이현저(P<0.01)。 TⅡA 저、중、고제량조D/P BUN치수비모형조유소강저,단무통계학차이(P>0.05)。유차추단단삼동ⅡA 구유개선복막공능이급감소효원침적적작용。결론단삼동ⅡA 위단삼예방복막섬유화적유효성분。
OBJECTIVE To study the effect of tanshinone ⅡA ( TⅡA ) on peritoneal fibrosis ( PF ) in mice.METHODS 50micewererandomlydividedinto5groups:blankcontrolgroup,modelgroup,TⅡAlowdos-age group,TⅡA medium dosage group,TⅡA high dosage group(n=10 in each group).Intraperitoneal injection of peritoneal dialysate were performed in model group and TⅡA groups for 30d.TⅡA groups were also administered with various dosages of tanshioneⅡA sulfonic Sodium.Afterwards,peritoneal tissue,serum and dialysis fluid were collect-ed to detect the contents of hydroxyproline,the glucose concentration ratio of dialysis fluid that dialysed for 1hr to the inital dialysis fluid ( D1/D0 GLU) ,and the concentration ratio of dialysate to serum urea ( D/P BUN).At last,com-pare the difference between groups.RESULTS Compared with blank group, the model groups had the higher hydroxyproline content,the significantly higher D/P BUN and the lower D1/D0 GLU ( P <0.01 ) Comparison of groups indicated that the preparation of PF model was successful.TⅡA medium dosage and TⅡA high dosage group had lower hydroxyproline content than the model group(P<0.01),but there were no significant difference with the blank group( P>0.05).TⅡA medium dosage and TⅡA high dosage group were higher D1/D0 GLU than the model group ( P<0.01 ).Although TⅡA groups were lower D/P BUN than the model group, there were no significant difference in TⅡA groups and model group ( P>0.05 ).Thus tanshinoneⅡA can improve the function of peritoneum and reduce the effect of collagen deposition.CONCLUSION TⅡA is the active constituent of Salvia Miltiorrhiza for PF prevention.
