目的:探讨2种方式移植人脐带间充质干细胞(UCMSC)对TNBS诱导的炎症性肠病小鼠模型的治疗作用。方法:分离培养人UCMSC,并通过一次性腹腔注射TNBS制备炎症性肠病动物模型,采用尾静脉注射和腹腔注射2种方式移植人UCMSC进行治疗,4周后处死,观察动物的死亡率、体重改变及结肠炎症变化,并进行大体和病理评分。结果:与对照组相比,尾静脉和腹腔注射移植UCMSC组动物死亡率下降,体重恢复较早,结肠病变大体评分和病理评分均显著改善;2种方式移植组之间的评分无显著差异。结论:尾静脉和腹腔移植人UCMSC对TNBS诱导的炎症性肠病小鼠模型均有治疗作用。

목적:탐토2충방식이식인제대간충질간세포(UCMSC)대TNBS유도적염증성장병소서모형적치료작용。방법:분리배양인UCMSC,병통과일차성복강주사TNBS제비염증성장병동물모형,채용미정맥주사화복강주사2충방식이식인UCMSC진행치료,4주후처사,관찰동물적사망솔、체중개변급결장염증변화,병진행대체화병리평분。결과:여대조조상비,미정맥화복강주사이식UCMSC조동물사망솔하강,체중회복교조,결장병변대체평분화병리평분균현저개선;2충방식이식조지간적평분무현저차이。결론:미정맥화복강이식인UCMSC대TNBS유도적염증성장병소서모형균유치료작용。
Objective: To compare the effect of tail vein injection and intraperitoneal injection of umbilical cord-derived mesenchymal stem cells(UCMSC) on the treatment of inflammatory bowel diseases in mice models. Meth?ods: The human UCMSC were isolated and cultured for use. The mice models of inflammatory bowel diseases were prepared by one intraperitoneal injection of TNBS, and 24 hours later, the UCMSC were injected from tail vein or intraperitoneally. After 4 weeks of treatment, the death rate and the weight recovery were recorded. The an?imals were executed and the colons were extracted for general observation and pathological evaluation. Results: Af?ter UCMSC treatment, either by tail vein injection or intraperitoneal injection, the animals' weight recovered earlier than control group, the death rate was lower. The evaluation of the colonal lesion improved in general observation as well as pathological detection, and the evaluation scores decreased statistically. However, the scores between the two different way injection groups have no significant difference. Conclusion: Transfusion of human UCMSC from tail vein or intraperitoneally are both effective in the treatment of inflammatory bowel diseases in mice mod?els.
