目的：检测健康人和乳腺癌患者外周血单个核细胞中多药耐药相关蛋白（ multidrug resistance－associated protein，MRP ）基因的表达水平，探讨MRP在乳腺癌患者中的可能作用。方法抽取20例健康人和50例乳腺癌患者外周血，运用ELISA法检测2组实验对象血清中肿瘤坏死因子α（TNF－α）、白介素－8（IL－8）、白介素6（IL－6）细胞因子的含量；提取外周血中单个核细胞（Peripheral blood monouclear cells ，PBMCs），后用荧光实时定量PCR和Western blot技术检测BCRP和MRP2／3／4／5 mRNA和相应蛋白表达情况。结果 ELISA法检测乳腺癌患者外周血血清中TNF－α、IL－8、IL－6细胞因子的含量明显高于健康人对照组（ P ＜0．05）；荧光实时定量PCR检测BCRP和MRP5的mRNA表达水平高于健康人对照组（ P ＜0．05），其余MRP没有显著性差别（ P ＞0．05）；Western blot 检测蛋白表达与mRNA 结果一致。结论乳腺癌患者外周血中的促炎因子含量的升高与BCRP和MRP5的表达调控有一定关系，具体相关机制有待进一步研究。
목적：검측건강인화유선암환자외주혈단개핵세포중다약내약상관단백（ multidrug resistance－associated protein，MRP ）기인적표체수평，탐토MRP재유선암환자중적가능작용。방법추취20례건강인화50례유선암환자외주혈，운용ELISA법검측2조실험대상혈청중종류배사인자α（TNF－α）、백개소－8（IL－8）、백개소6（IL－6）세포인자적함량；제취외주혈중단개핵세포（Peripheral blood monouclear cells ，PBMCs），후용형광실시정량PCR화Western blot기술검측BCRP화MRP2／3／4／5 mRNA화상응단백표체정황。결과 ELISA법검측유선암환자외주혈혈청중TNF－α、IL－8、IL－6세포인자적함량명현고우건강인대조조（ P ＜0．05）；형광실시정량PCR검측BCRP화MRP5적mRNA표체수평고우건강인대조조（ P ＜0．05），기여MRP몰유현저성차별（ P ＞0．05）；Western blot 검측단백표체여mRNA 결과일치。결론유선암환자외주혈중적촉염인자함량적승고여BCRP화MRP5적표체조공유일정관계，구체상관궤제유대진일보연구。
Objective To investigate the expression of multidrug resistance-associated protein ( MRP) in peripheral blood monouclear cells ( PBMCs) of healthy people and patients with breast cancer ,and to explore their possible roles in the pathogenesis of breast cancer .Methods The levels of TNF-α,IL-8 and IL-6 in serum were detected by ELISA in 50 patients with breast cancer ( patients group ) and 20 healthy subjects ( control group ) .After the PBMCs in both groups were extracted , the expression levels of breast cancer resistance protein ( BCRP ) and MRP2/3/4/5 mRNA were detected by Real-time quantitative RT-PCR and Western blotting .Results The levels of TNF-α,IL-8 and IL-6 in patients group were significantly higher than those in control group ( P <0.05 ).The expression levels of BCRP and MRP5 mRNA were significantly increased,as compared with those in control group ( P <0.05),however,there were no significant differences in the other kinds of MRP between two groups ( P >0.05).Conclusion The increase of proinflammatory factors in peripheral blood of patients with breast cancer may be correlated to the expressions of BCRP and MRP 5,however,the related mechanism needs to be studied further .
