以棉花抗黄萎病品系苏抗045和感黄萎病品系苏研116为亲本构建了一个含有237个F2单株的作图群体.利用中等致病力黄萎病菌株BP2对P1、P2、F1、F2:3家系群体进行纸钵撕底菌液醮根鉴定黄萎病抗性,计算各世代的相对病指.用2400对SSR引物进行筛选,获得78个SSR多态性标记位点,进一步利用JoinMap作图软件,构建了一张陆陆杂种遗传连锁图,包含41个标记、15个连锁群,总长为359.90 cM,覆盖棉花总基因组约7.2%.利用复合区间作图法分析,在NAU2970~MUSS138区间内检测到1个抗黄萎病QTL,可解释的表型变异为9.69%.

Up1and cotton Sukang045, resistant to Verticillium wi1t, and the susceptib1e variety, Suyan116, were used as parenta1 materia1s to deve1op a tagging popu1ation with 237 F2 individua1 p1ants. By inocu1ating pathogenic fungus strain BP2 to the popu1ations of P1 , P2 , F1 , F2 : 3 , the re1ative disease indexes of each generation were scored. A tota1 of 2 400 pairs of SSR primers were screened, and 78 were po1ymorphic. Using JoinMap software, a genetic 1inkage map of in-traspecific F2 popu1ations was constructed. The 1inkage map was comprised of 41 1oci on 15 1inkage groups, covering 359. 90 cM and approximate1y 7. 2% of the tota1 cotton genome. One resistance QTL was detected and 1oca1ized in the in-terva1 of NAU2970 and MUSS138, responsib1e for 9. 69% phenotypic variance in F2 : 3 .
