为了解江苏淮北小麦品种重要品质性状相关基因状况,以20份近期审定的徐麦系列和淮麦系列小麦品种以及12份小麦对照材料为试验材料,采用SDS-PAGE电泳与PCR检测相结合,检测了小麦高分子量麦谷蛋白亚基;同时采用共显性PCR标记筛查1BL/1RS易位系以及Wx基因突变型。结果显示,江苏淮北小麦品种Glu-A1位点含3种亚基,其中1亚基占75%;Glu-B1位点有4种亚基,7+8亚基占60%;Glu-D1位点有3种亚基,5+10亚基占50%;1BL/1RS易位系有14份,占全部品种的70%;所有品种均为Wx基因野生型。

Twenty commercia1 varieties authorized recent1y, inc1uding 9 Xumai varieties and 11 Huaimai varieties, and 12 contro1 varieties were se1ected as materia1s to identify the composition of high mo1ecu1ar weight g1utenin subunits (HMW-GS) by SDS-PAGE and PCR. The Wx mutant genes and 1BL/1RS trans1ocation 1ine were screened by PCR using co-dominant markers. Three subunit types were discovered at 1ocus Glu-A1, subunit 1 accounting for 75%. There were four subunit types at 1ocus Glu-B1, subunit 7+8 accounting for 60%. Three subunit types were identified at 1ocus Glu-D1, sub-unit 5+10 accounting for 50%. Fourteen varieties with 1BL/1RS trans1ocation were discovered, most of 1ohich (70%) came from Huaimai varieties. No Wx mutant gene was detected in a11 tested Huaibei varieties.
