为阐明番茄抗根结线虫基因Mi-1在土壤温度高于28益时丧失抗性的原因,选用3份番茄材料Mote11e (Mi-1)、LA2157(Mi-9)和新选育获得的具有热稳定抗根结线虫特性的栽培番茄材料ZN17(Mi-HT)进行研究。通过根结线虫的人工接种鉴定,确定Mote11e在高温条件下丧失抗性,而ZN17和LA2157在高温条件下仍然具有良好的抗性。利用特异引物对各番茄材料的cDNA模板进行扩增、测序和序列比对分析,发现这3份材料扩增片段具有很高的相似性,因此利用该引物对各材料高温和常温条件下的基因表达变化进行分析,结果表明,Mi-1在高温条件下不表达是番茄材料Mote11e高温条件下对根结线虫抗性丧失的原因。番茄材料LA2157和ZN17中扩增得到的相关基因表达状况与其在高温条件下对根结线虫的抗性表现相吻合。

To identify the reason of resistanc1oss of tomato to root-knot nematode when the soi1 temperature is above 28℃, tomato 1ines Mote11e(Mi-1), LA2157(Mi-9)and a new bred cu1tivated tomato 1ine ZN17(Mi-HT) with the heat-stab1e root-knot nematode resistance were chosen in this research. After nematode artificia1 inocu1ation, the nematode resistance of Mote11e was detected inactive, whi1e the resistances of ZN17 and LA2157 were active at high temperature. Comp1ementary DNA of tomato 1ines were used as the temp1ate to conduct po1ymerase chain reaction with specific primer. The amp1ified prod-ucts were sequenced, and BLAST ana1ysis was performed. The resu1t showed that the sequences of amp1ified fragment shared high simi1arity. So the specific primer was emp1oyed to ana1yze the expression of re1ated gene S, Mi-1, Mi-9 and Mi-HT. Anay1is revea1ed that the si1ence of Mi-1 was responsib1e for 1ose resistance 1oss in Mote11e at high temperature. The expres-sions of homo1ogous gene fragments amp1ified from LA2157 and ZN17 with the Mi-1 primer at high temperature was observed to be consistent with the resistance performance to tomato root-knot nematode.
