以陆地棉杂交组合DPLSP3×盐城115(群体A)和Humcopedixi14wtr5×盐城90-110(群体B)构建的2个陆地棉种内F2群体为材料,发掘稳定的棉籽油分含量及蛋白质含量QTL,为标记辅助选择提供依据.利用覆盖棉花基因组的SSR标记对陆地棉油分和蛋白质含量数量性状位点进行筛选.基于该群体,以2009和2010年油分和蛋白质含量为指标,复合区间作图分析结果表明,2年2个群体共检测到4个QTL.其中,在群体A中,定位到1个较稳定的控制油分含量的QTL,该QTL位于染色体13上DC20120-NAU2893标记区间,在F2及F2:3群体中分别可以解释9.21%和12.01%的遗传变异;另外,在F3世代检测到1个蛋白质含量QTL.在群体B的F3世代,检测到1个位于NAU2932-NAU2503标记区间的油分含量QTL和1个蛋白质含量QTL,分别可以解释24.67%和10.1%的遗传变异.这些稳定遗传的QTL 可以用于辅助陆地棉品质改良.

In this study, two F2 popu1ations from the intraspecific cross DPLSP3×Yancheng115 (A) and Humcope-dixi14wtr5×Yancheng90-110 (B) were used as experimenta1 materia1s to identify stab1e QTLs associated with cottonseed oi1 and protein contents, and to provide information for mo1ecu1ar marker-assisted breeding. SSR primers covering the who1e cotton genome were se1ected to detect the QTLs in 2009 and 2010 in up1and cotton. A tota1 of 4 QTLs contro11ing cottonseed oi1 and protein contents were identified by the composite interva1 mapping. In popu1ation A, a stab1e QTL for oi1 content was identified between the marker interva1 DC20120 and NAU2893 on chr. 13, exp1aining 9. 21% and 12. 01% of the tota1 variation in F2 and F2:3 generations, respective1y. Another QTL for protein content was detected between NAU3377 and NAU5192 on chr. 11 on1y in F3 generation, exp1aining 9. 85% of phenotypic variation. In popu1ation B, a significant QTL qOC-19-1 for oi1 content and a QTL qPC-19-1 for protein content were mapped between the marker NAU2932 and NAU2503, exp1aining 24. 67% and 10. 1% of the phenotypic variations in F3 generation. Humcopedixi14wtr5 and Yancheng90-110 were genotyped as increased oi1 content and increased protein content, respective1y. The stab1y heritab1e QTL cou1d be used for the mo1ecu1ar breeding of cottonseed nutrient traits in up1and cotton.
