本研究对一个叶夹角变小、叶片短而直立的突变体ser( Short and erect)进行了植物激素敏感性试验和基因定位.激素试验结果表明,ser对赤霉素( Gibbere11ic acid, GA)敏感而对油菜素内酯( Brassino1ide, BR)敏感性降低.遗传分析结果表明,ser的直立叶性状受1对隐性单基因ser控制.在ser×08CR578 F2群体中,利用分子标记将ser基因定位在水稻第5染色体长臂上的标记RM3437与RM5454之间,ser基因距离2个标记的遗传距离分别为5.3 cM和1.5 cM;在ser×L604 F2群体中,ser基因定位于第5染色体上标记RM18504与RM1237之间,距离2个标记的遗传距离分别为1.5 cM和2.1 cM.在2个群体中,ser基因均与标记RM18532共分离.本研究为进一步精细定位与克隆ser基因奠定了基础,也为解析水稻叶夹角变化机制及选育高产理想株型水稻提供了理论支撑和基因资源.

A short and erect rice (ser) mutant was used for phytohormone sensitivity ana1ysis and genetic mapping. Phy-tohormone treatment showed that the mutant was sensitive to gibbere11ic acid ( GA) and insensitive to brassino1ide ( BR) . Genetic ana1ysis revea1ed that the erect 1eaf was governed by a recessive nuc1eus gene, ser. The ser gene was first1y identified with 500 pairs of simp1e sequence repeat (SSR) markers covering the who1e genome by BSA with F2 popu1ations derived from F1s of ser× 08CR578 and ser×L604. In F2 popu1ation of ser×08CR578, ser gene was 1ocated on the 1ong arm of chromosome 5 between RM3437 and RM5454, with genetic distances of 5. 3 cM and 1. 5 cM respective1y, and co-segregated with RM18532. In F2 popu-1ation of ser×L604, ser gene was 1ocated on the 1ong arm of chromosome 5 between RM18504 and RM1237, with genetic distances of 1. 5 cM and 2. 1 cM, respective1y, and co-segregated with RM18532 as we11. The study 1aid the foundation for further fine-mapping and c1oning of the gene, and wou1d contribute to the defining of regu1ation mechanism of 1eaf ang1e and supp1y a genetic resource for breeding ideotype rice.
