目的:利用生物信息学预测胃癌中 P53诱导相关微小 RNA(miRNA)的靶基因及功能。方法通过胃癌的限定,用在线数据库mir2disease、Gene Expression Atlas缩小目的基因范围。利用在线预测软件miRNA 靶基因数据库 miRGen(v3.0)预测目标miRNA的靶基因。通过在线软件DAVID和Pathway Miner对预测的靶基因数据集进行功能富集和信号转导通路分析。结果胃癌中P53诱导相关miRNA包括miR-21、miR-25、miR-103等,预测miR-21靶基因共1090个,其中在胃癌中下调共115个;Pathway Miner分析显示miR-21的靶基因涉及黏附连接、胰岛素信号转导通路、细胞凋亡、钙信号转导通路、JAK-STAT信号转导途径等。结论 miR-21靶基因涉及多个肿瘤发生、发展相关信号通路。

목적:이용생물신식학예측위암중 P53유도상관미소 RNA(miRNA)적파기인급공능。방법통과위암적한정,용재선수거고mir2disease、Gene Expression Atlas축소목적기인범위。이용재선예측연건miRNA 파기인수거고 miRGen(v3.0)예측목표miRNA적파기인。통과재선연건DAVID화Pathway Miner대예측적파기인수거집진행공능부집화신호전도통로분석。결과위암중P53유도상관miRNA포괄miR-21、miR-25、miR-103등,예측miR-21파기인공1090개,기중재위암중하조공115개;Pathway Miner분석현시miR-21적파기인섭급점부련접、이도소신호전도통로、세포조망、개신호전도통로、JAK-STAT신호전도도경등。결론 miR-21파기인섭급다개종류발생、발전상관신호통로。
Objective To predict target genes and functions of P53-induced miRNAs in gastric cancer by bioinformatics analysis. Methods Databases of mir2disease and Gene Expression Atlas were used for extraction P53-induced miRNAs in gastric cancer from published literature.The target genes of related miRNA were predicted by miRGen(v3.0)Database.Then DAVID and Path-way Miner were used for Gene Ontology Enrichment and pathway analysis of the target genes.Results P53 induced miRNAs in human gastric cancer included miR-21、miR-25、miR-103.Among them,1 090 predicted target genes of miR-21 were found in total and 1 1 5 in gastric cancer,respectively.The pathway analysis showed that these target genes were involved in Adherens j unction, Adipocytokine signaling pathway,apoptosis,calcium signaling pathway and JAK-STAT signaling pathway,etc.Conclusion Bioin-formatics analysis suggests P53 induced miR-21 expression in gastric cancer,and its target genes are involve in multiple signaling pathways which related tumor pathogenesis and progression.
