目的:探究霍奇金淋巴瘤(HL)细胞SH2结构域酪氨酸磷酸化酶(SHP1)表达与CD99表达的关系。方法荧光定量PCR(RT‐PCR)、Western blot检测 HL淋巴瘤细胞株L428细胞、B淋巴母细胞株IM9细胞、浆母细胞株KM3细胞及上调CD99的稳定细胞株L428‐CD99细胞中CD99和SHP1 mRNA、蛋白水平的表达;荧光共聚焦观测L428细胞中CD99与SHP1的表达及其共定位;IM9细胞瞬时干扰CD99在基因和蛋白水平检测SHP1的表达。结果 L428和KM3细胞CD99和SHP1 mR‐NA呈现低表达,L428‐CD99细胞SHP1基因和蛋白水平较L428细胞表达增高,其蛋白表达与基因转录水平表达趋势相一致;随着CD99的上调,SHP1基因和蛋白水平表达升高;CD99为细胞膜表达,SHP1为细胞质表达;瞬时干扰CD99后,SHP1在基因和蛋白水平表达均降低。结论 SHP1在 H L中低表达可能与其CD99缺失有关,其机制仍有待于研究。
目的:探究霍奇金淋巴瘤(HL)細胞SH2結構域酪氨痠燐痠化酶(SHP1)錶達與CD99錶達的關繫。方法熒光定量PCR(RT‐PCR)、Western blot檢測 HL淋巴瘤細胞株L428細胞、B淋巴母細胞株IM9細胞、漿母細胞株KM3細胞及上調CD99的穩定細胞株L428‐CD99細胞中CD99和SHP1 mRNA、蛋白水平的錶達;熒光共聚焦觀測L428細胞中CD99與SHP1的錶達及其共定位;IM9細胞瞬時榦擾CD99在基因和蛋白水平檢測SHP1的錶達。結果 L428和KM3細胞CD99和SHP1 mR‐NA呈現低錶達,L428‐CD99細胞SHP1基因和蛋白水平較L428細胞錶達增高,其蛋白錶達與基因轉錄水平錶達趨勢相一緻;隨著CD99的上調,SHP1基因和蛋白水平錶達升高;CD99為細胞膜錶達,SHP1為細胞質錶達;瞬時榦擾CD99後,SHP1在基因和蛋白水平錶達均降低。結論 SHP1在 H L中低錶達可能與其CD99缺失有關,其機製仍有待于研究。
목적:탐구곽기금림파류(HL)세포SH2결구역락안산린산화매(SHP1)표체여CD99표체적관계。방법형광정량PCR(RT‐PCR)、Western blot검측 HL림파류세포주L428세포、B림파모세포주IM9세포、장모세포주KM3세포급상조CD99적은정세포주L428‐CD99세포중CD99화SHP1 mRNA、단백수평적표체;형광공취초관측L428세포중CD99여SHP1적표체급기공정위;IM9세포순시간우CD99재기인화단백수평검측SHP1적표체。결과 L428화KM3세포CD99화SHP1 mR‐NA정현저표체,L428‐CD99세포SHP1기인화단백수평교L428세포표체증고,기단백표체여기인전록수평표체추세상일치;수착CD99적상조,SHP1기인화단백수평표체승고;CD99위세포막표체,SHP1위세포질표체;순시간우CD99후,SHP1재기인화단백수평표체균강저。결론 SHP1재 H L중저표체가능여기CD99결실유관,기궤제잉유대우연구。
Objective To explore the relationship between SHP1 and CD99 expression in Hodgkin lymphoma (HL) .Methods RT‐PCR and Western blot to detect the expression of CD99 and SHP1 mRNA and protein expression in IM9、KM3、L428、L428‐CD99 cells ;fluorescence confocal to observe the expression and co‐localization of CD99 and SHP1 ;transiently interference CD99 in IM9 cells and then detect the expressing of SHP1 mRNA and protein levels .Results SHP1 mRNA was low expressed in L428、KM3 and high expressed in IM9;gene and protein expression were consistent trend;in L428‐CD99 cell SHP1 mRNA expression and protein levels were higher than L428 cell;CD99 expressed in membrane and SHP1 expressed in cell plasma;transient interference CD99 ,SHP1 decreased in mRNA expression and protein levels .Conclusion In HL cells ,SHP1 expression related to the missing expression of CD99 ,and its mechanism remains to be studied .
