目的：研究银丹心脑通软胶囊（YXC）对血管内皮细胞的保护作用及作用机制。方法采用氧化低密度脂蛋白（Ox-LDL）和肿瘤坏死因子α（TNF-α）诱导人脐静脉内皮细胞（HUVEC）损伤两种模型，MTT 法测定 YXC 预处理后 HUVEC 存活率，探讨 YXC 对内皮细胞的保护作用；采用流式细胞术测定 YXC 对内皮细胞凋亡的影响，探讨YXC 内皮保护的作用机制。结果 YXC 的最大无毒浓度为500μg·mL-1。将250、125、62.5μg·mL-13个浓度作为YXC 的实验浓度；YXC 孵育24 h 后可明显升高 Ox-LDL 诱导的 HUVEC 的存活率（P <0.05），具有明显的量效关系；YXC 孵育24 h 后可明显升高 TNF-α诱导的 HUVEC 的存活率（P <0.05）。250和125μg·mL-1YXC 可显著降低HUVEC 细胞的凋亡率。结论 YXC 对血管内皮细胞具有保护作用，其机制可能与抗细胞凋亡有关。
목적：연구은단심뇌통연효낭（YXC）대혈관내피세포적보호작용급작용궤제。방법채용양화저밀도지단백（Ox-LDL）화종류배사인자α（TNF-α）유도인제정맥내피세포（HUVEC）손상량충모형，MTT 법측정 YXC 예처리후 HUVEC 존활솔，탐토 YXC 대내피세포적보호작용；채용류식세포술측정 YXC 대내피세포조망적영향，탐토YXC 내피보호적작용궤제。결과 YXC 적최대무독농도위500μg·mL-1。장250、125、62.5μg·mL-13개농도작위YXC 적실험농도；YXC 부육24 h 후가명현승고 Ox-LDL 유도적 HUVEC 적존활솔（P <0.05），구유명현적량효관계；YXC 부육24 h 후가명현승고 TNF-α유도적 HUVEC 적존활솔（P <0.05）。250화125μg·mL-1YXC 가현저강저HUVEC 세포적조망솔。결론 YXC 대혈관내피세포구유보호작용，기궤제가능여항세포조망유관。
Objective To study the protective effect of Yindan Xinnaotong capsule (YXC) on endothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were cultured in vitro and HUVECs were pretreated with 250 μg·mL-1, 125 μg·mL-1 and 62.5 μg·mL-1 for 24 h. 200 μg·mL-1 Ox-LDL and 60 ng·mL-1 TNF-α were used to induce HUVECs injury. The cell survival rate was determined by MTT, and flow cytometry was used to detect apoptosis to evaluate the mechanism of protective function of YXC. Results The maximal non-cytotoxic concentration of YXC was 500 μg·mL-1, thus 250, 125 and 62.5 μg·mL-1 were employed as the experimental concentrations. YXC could obviously increase the survival rate of HUVECs induced by Ox-LDL (P <0.05) and TNF-α with a dose-dependence manner; compared with model group, 62.5 and 250 μg·mL-1 YXC treatment decreased apoptosis rate induced by Ox-LDL HUVECs (P <0.05). Meanwhile, 250 and 125 μg·mL-1 YXC obviously reduced apoptosis rate induced by TNF-α(P <0.05). Conclusion YXC could protect the HUVEC injury induced by Ox-LDL and TNF-α, and this may be associated with the inhibitory effect on cell apoptosis.
