噬菌体展示技术是一种特殊的基因工程重组表达技术,已经作为一种强大的工具运用在不同的研究中,包括筛选抗体和酶的配体、筛选小分子的受体、基因工程抗体等。免疫分析作为农药残留快速筛查技术被广泛研究,利用抗体从噬菌体展示肽库中淘选竞争物,可以加快异源免疫分析方法的建立;利用抗原抗体复合物从噬菌体展示肽库中淘选抗免疫复合体多肽,可建立非竞争免疫分析方法,丰富农药的免疫分析模式。本文从噬菌体展示技术的原理、噬菌体展示肽库的构建和噬菌体展示肽库的筛选方面概述了噬菌体展示肽库技术;从模拟表位筛选、抗免疫复合体筛选和新免疫分析方法方面阐述了噬菌体展示肽库技术在农药等小分子免疫分析中的应用;并对噬菌体展示肽库技术的研究和应用前景进行了展望。

Phage display technology (PDT) is a unique genetic engineering recombinant expression technology, which is a powerful tool for a variety of applications, including antibody engineering and the isolation of peptide ligands for antibodies and enzymes and peptide receptors for small molecules. Immunoassay had been widely studied as a rapid screening technique for pesticide residue, isolation of competitor from phage display peptide library by using antibody could accelerate the development of heterologous immunoassay; isolation of anti-immunocomplex peptide from phage display peptide library by using antigen-antibody complexes could develop noncompetitive immunoassay, and enriched immunoassay model for pesticide. This review concisely summarized phage display peptide library technology from the principle of phage display technology, the construction of phage display peptide library and screening of phage display peptide library. Appling phage display peptide library technology in immunoassay for pesticides and other small molecules were elaborated from screening of mimicking epitope, screening of anti-immunocomplex and the new immunoassays. Furthermore, this review presented insights for the study and application prospect of the phage display peptide library technology.
