目的:对高效液相色谱法测定水产品(鲭鱼)中组胺含量的试验方法进行改进,建立一种提取方法简单、衍生物稳定、操作方便的检测方法。方法用0.4 mol/L高氯酸溶液提取水产品中的组胺,采用10 mg/mL丹磺酰氯溶液作为衍生剂进行衍生,用甲醇、水的流动相梯度洗脱,高效液相色谱-紫外检测法测定水产品中的组胺含量。结果该方法提取完全、操作简单、峰型理想、组胺出峰干扰少,回收率大于90%,方法检出限为50 mg/kg。结论该方法能够满足水产品中组胺含量的测定要求,是一种理想的组胺检测方法。
目的:對高效液相色譜法測定水產品(鯖魚)中組胺含量的試驗方法進行改進,建立一種提取方法簡單、衍生物穩定、操作方便的檢測方法。方法用0.4 mol/L高氯痠溶液提取水產品中的組胺,採用10 mg/mL丹磺酰氯溶液作為衍生劑進行衍生,用甲醇、水的流動相梯度洗脫,高效液相色譜-紫外檢測法測定水產品中的組胺含量。結果該方法提取完全、操作簡單、峰型理想、組胺齣峰榦擾少,迴收率大于90%,方法檢齣限為50 mg/kg。結論該方法能夠滿足水產品中組胺含量的測定要求,是一種理想的組胺檢測方法。
목적:대고효액상색보법측정수산품(청어)중조알함량적시험방법진행개진,건립일충제취방법간단、연생물은정、조작방편적검측방법。방법용0.4 mol/L고록산용액제취수산품중적조알,채용10 mg/mL단광선록용액작위연생제진행연생,용갑순、수적류동상제도세탈,고효액상색보-자외검측법측정수산품중적조알함량。결과해방법제취완전、조작간단、봉형이상、조알출봉간우소,회수솔대우90%,방법검출한위50 mg/kg。결론해방법능구만족수산품중조알함량적측정요구,시일충이상적조알검측방법。
Objective To improve the method of high performance liquid chromatography (HPLC) for determination of histamine in aquatic product (mackerel fish), and to develop a method with simple extraction, stable derivatives, and easy to operate.Method The perchloric acid solution 0.4 mol/L was used to extract histamine of aquatic products, dansyl chloride solution 10 mg/mL as derivative agent, methanol and water as the HPLC flow phase to gradient elution. The histamine content was detected by high performance liquid chromatography with ultraviolet detector.Results The method had the advantages of complete extraction, simple operation, ideal peak, less interference of histamine peak. The recovery rate was more than 90%, and the detection limit was 50 mg/kg.Conclusion It can meet the requirements of determination of histamine in aquatic products, and is an ideal method for detection of histamine.
