目的:建立日本血吸虫重叠 HBV 感染小鼠模型,观察其生物学表现。方法日本血吸虫尾蚴感染6~8周龄雌性 BALB/cJ 小鼠,8周后,通过高压尾静脉注射 B 基因型 HBV 可复制性克隆,分别在注射后第3、7天,检测血清HBV DNA、HBsAg、HBeAg、ALT、透明质酸(HA)水平;检测 HBcAg 在肝细胞内表达情况和虫卵肉芽肿形成情况。比较重叠感染相关生物学指标和单独感染组的不同。结果重叠感染组小鼠肝组织内可见 HBcAg 在虫卵肉芽肿周围肝细胞内表达;可以正常分泌 HBsAg 和 HBeAg 以及子代病毒;其血清 ALT 水平低于 HBV 感染组,高于血吸虫感染组;HA 含量相对明显升高。结论成功建立了日本血吸虫重叠 HBV 感染小鼠模型,为后续研究此类重叠感染提供了良好的小动物实验平台。
目的:建立日本血吸蟲重疊 HBV 感染小鼠模型,觀察其生物學錶現。方法日本血吸蟲尾蚴感染6~8週齡雌性 BALB/cJ 小鼠,8週後,通過高壓尾靜脈註射 B 基因型 HBV 可複製性剋隆,分彆在註射後第3、7天,檢測血清HBV DNA、HBsAg、HBeAg、ALT、透明質痠(HA)水平;檢測 HBcAg 在肝細胞內錶達情況和蟲卵肉芽腫形成情況。比較重疊感染相關生物學指標和單獨感染組的不同。結果重疊感染組小鼠肝組織內可見 HBcAg 在蟲卵肉芽腫週圍肝細胞內錶達;可以正常分泌 HBsAg 和 HBeAg 以及子代病毒;其血清 ALT 水平低于 HBV 感染組,高于血吸蟲感染組;HA 含量相對明顯升高。結論成功建立瞭日本血吸蟲重疊 HBV 感染小鼠模型,為後續研究此類重疊感染提供瞭良好的小動物實驗平檯。
목적:건립일본혈흡충중첩 HBV 감염소서모형,관찰기생물학표현。방법일본혈흡충미유감염6~8주령자성 BALB/cJ 소서,8주후,통과고압미정맥주사 B 기인형 HBV 가복제성극륭,분별재주사후제3、7천,검측혈청HBV DNA、HBsAg、HBeAg、ALT、투명질산(HA)수평;검측 HBcAg 재간세포내표체정황화충란육아종형성정황。비교중첩감염상관생물학지표화단독감염조적불동。결과중첩감염조소서간조직내가견 HBcAg 재충란육아종주위간세포내표체;가이정상분비 HBsAg 화 HBeAg 이급자대병독;기혈청 ALT 수평저우 HBV 감염조,고우혈흡충감염조;HA 함량상대명현승고。결론성공건립료일본혈흡충중첩 HBV 감염소서모형,위후속연구차류중첩감염제공료량호적소동물실험평태。
Objective To establish a mouse model of Schistosoma japonicum and hepatitis B virus (HBV)co-infection.Methods Female BAL B/cJ mice of 6-8 weeks old were infected by cercariae of Schistosoma japonicumat.In 8 weeks,HBV replication competent plasmid of genotype B was injected to these mice through high-pressure tail intravenous. Then Blood samples were collected at day 3 and 7,respectively.Levels of HBsAg,HBeAg,ALT,HA,and HBV titer were detected. In addition, HBcAg expression in hepatocyte and formation of egg-granuloma were examined by immunohistochemical staining and HE staining,respectively.Related biological indexes were compared between different groups.Results HBcAg expresssion was detected in hepatocytes around ova granuloma in liver tissue of super-infection mice.HBsAg,HBeAg and progeny virus were also detected in mice serum.ALT level in super-infection mice was lower than that of merely HBV-infected mice,but higher than that of group infected with Schistosoma japonicum alone.While HA level was significantly higher than that of mice infected by HBV or Schistosoma japonicum,respectively.Conclusion The mice model with super-infection of Schistosoma japonicum and HBV was constructed successfully.It could provide an experimental platform of small animal for subsequent research associated with Schistosoma japonicum and HBV superinfection.
