目的:研究 MexXY-OprM主动外排系统在铜绿假单胞菌(PA)多重耐药机制中的作用。方法挑选临床分离的PA 多重耐药菌株26株、敏感菌株15株及标准质控菌株(ATCC27853),用 RT-PCR 方法扩增其内膜转运蛋白 MexY 的结构基因mexY 和内参基因16SrRNA 片段,根据结构基因 mexY 与16SrRNA 扩增产物的电泳扫描比值,分析 mexY mRNA 的表达水平,用以判断 MexXY-OprM的表达情况。结果26株多重耐药菌株对14种常用抗菌药物都耐药,耐药率为100.00%,15株敏感菌株对14种常用抗菌药物都敏感;标准质控菌株、敏感菌株和多重耐药菌株均检测出 mexY 的 mRNA 基因,耐药菌株的 mexY mRNA 水平高于敏感菌株(P <0.05)。结论MexXY-OprM主动外排系统表达水平的增高在 PA 多重耐药中起到重要的作用。

목적:연구 MexXY-OprM주동외배계통재동록가단포균(PA)다중내약궤제중적작용。방법도선림상분리적PA 다중내약균주26주、민감균주15주급표준질공균주(ATCC27853),용 RT-PCR 방법확증기내막전운단백 MexY 적결구기인mexY 화내삼기인16SrRNA 편단,근거결구기인 mexY 여16SrRNA 확증산물적전영소묘비치,분석 mexY mRNA 적표체수평,용이판단 MexXY-OprM적표체정황。결과26주다중내약균주대14충상용항균약물도내약,내약솔위100.00%,15주민감균주대14충상용항균약물도민감;표준질공균주、민감균주화다중내약균주균검측출 mexY 적 mRNA 기인,내약균주적 mexY mRNA 수평고우민감균주(P <0.05)。결론MexXY-OprM주동외배계통표체수평적증고재 PA 다중내약중기도중요적작용。
Objective To investigate the effect of MexXY-OprMactive efflux system on pseudomonas aeruginosa.Methods Twen-ty-six strains of multiple drug resistant strains of pseudomonas aeruginosa and 15 strains of sensitive strains and standard quality control strains were selected.The levels of mexY mRNA of 15 sensitive strains and 26 multiple drug resistant strains were detected by RT-PCR.The 16SrRNA gene was also amplified as control,and the levels of mexY mRNA were analyzed according to the ratio of electrophoresis scan results of their PCR products and the 16SrRNAs.Results Twenty-six strains of multiple drug resistant strains showed resistant to 14 kinds of com-monly used antibiotics.Fifteen strains of sensitive strains showed sensitive to 14 kinds of commonly used antibiotics.mexY mRNA was detec-ted in sensitive strains,standard quality control strains and multiple drug resistant strains.The levels of mexY mRNA in drug-resistant strains were higher than in susceptible strains(P <0.05 ).Conclusion In multidrug resistant pseudomonas aeruginosa,the over-expression of MexXY-OprMplays an important role.
