目的:探讨MicroRNA-100(miR-100)在胃癌患者血清中的表达及临床意义。方法:选择蚌埠医学院第一附属医院手术及随访资料完整的胃癌患者(40例)和健康对照者(40例)为研究对象,提取血清总miRNA,在建立了稳定、敏感的血清miR-100绝对定量检测方法(qRT-PCR)的基础上,检测胃癌和健康对照者血清中 miR-100表达水平。分析胃癌和健康对照者血清中miR-100表达差异及血清中 miR-100表达水平与胃癌临床病理参数的关系。结果:胃癌患者血清中 miR-100的表达水平为(2.78±1.92)fmol/L,显著高于健康对照者[(0.19±0.15)fmol/L,P<0.01],同时miR-100的受试者工作特征曲线显示,血清miR-100对胃癌诊断具有良好的特异度和敏感度(曲线下面积0.985);进一步分析发现:血清中 miR-100的表达与性别、年龄、肿瘤直径大小、浸润深度、分化程度、淋巴结转移数量、TNM分期等无明显差异(P>0.05)。结论:血清miR-100的检测可能有助于胃癌的诊断。

목적:탐토MicroRNA-100(miR-100)재위암환자혈청중적표체급림상의의。방법:선택방부의학원제일부속의원수술급수방자료완정적위암환자(40례)화건강대조자(40례)위연구대상,제취혈청총miRNA,재건립료은정、민감적혈청miR-100절대정량검측방법(qRT-PCR)적기출상,검측위암화건강대조자혈청중 miR-100표체수평。분석위암화건강대조자혈청중miR-100표체차이급혈청중 miR-100표체수평여위암림상병리삼수적관계。결과:위암환자혈청중 miR-100적표체수평위(2.78±1.92)fmol/L,현저고우건강대조자[(0.19±0.15)fmol/L,P<0.01],동시miR-100적수시자공작특정곡선현시,혈청miR-100대위암진단구유량호적특이도화민감도(곡선하면적0.985);진일보분석발현:혈청중 miR-100적표체여성별、년령、종류직경대소、침윤심도、분화정도、림파결전이수량、TNM분기등무명현차이(P>0.05)。결론:혈청miR-100적검측가능유조우위암적진단。
Objective: To investigate the expression of microRNA-100 (miR-100) in the serum of gastric cancer patients and its clinical significance. Methods: At The First Affiliated Hospital of Bengbu Medical College, 40 gastric cancer patients who underwent surgery with complete follow-up data and 40 healthy controls were selected as research subjects. Total miRNA was isolated from the se-rum samples of the cancer patients. A stable and sensitive detection method for the absolute quantification of miR-100 was established. The serum levels of miR-100 in the patients and healthy controls were tested according to this novel method. Differences of the miR-100 expression in the serum samples of the patients and healthy controls were analyzed using statistical analysis. The correlation between miR-100 expression levels and the clinicopathological features of gastric cancer was also analyzed. Results: The expression level of miR-100 in the serum was significantly higher in cancer patients[(2.78±1.92) fmol/L]than in the healthy controls[(0.19± 0.15) fmol/L, P<0.01]. The receiver operating characteristic curve of miR-100 indicated that serum miR-100 has satisfactory specificity and sensitivity for the diagnosis of gastric cancer (AUC=0.985). Further analytical results indicated that no significant differences were found among the expression levels of serum miR-100 with respect to the gender, age, tumor diameter, infiltrating depth, differentiation degree, frequency of lymph-node metastasis, and TNM stage (P>0.05). Conclusion: Serum miR-100 testing may be helpful in the diag-nosis of gastric cancer.
