目的:观察麝香乌龙丸对糖尿病( DM)兔骨折愈合的影响,并探讨其机制。方法将36只大白兔随机分为A、B、C组各12只。 B、C组建立DM模型,3组均造成右胫腓骨骨折。 C组将麝香乌龙丸以1.0 g/kg剂量溶于5 mL生理盐水中灌胃,A、B组给予等量生理盐水灌胃。3组分别于给药后第1、2、4周各取4只大白兔,麻醉后采用Norland.XR 36双能X线骨密度测量仪检测骨折端骨密度,取骨痂行HE染色观察形态学变化,采用Western blot法检测骨痂神经生长因子( NGF)相对表达。结果 B组局部骨密度在骨折愈合过程中出现下降,给药1~2周时略微下降,第4周下降明显,其下降趋势较A、C组更加明显。 B组各时点骨密度均低于A、C组,P均<0.05。3组用药后第1周的骨痂形态学均无明显变化;用药第2、4周与B组比较,A组可见大量较成熟软骨存在,形成软骨岛,骨小梁生成并发育成熟;C组未见软骨岛存在,但软骨性骨痂加速向骨性骨痂转化。 B、C组用药第2、4周的骨痂NGF相对表达均较第1周升高,B组各时点骨痂NGF相对表达均低于A、C组,P均<0.05;A、C组各时点骨痂NGF相对表达比较,P均>0.05。结论麝香乌龙丸可以加速DM兔骨折愈合,可能与上调骨痂NGF的表达有关。
目的:觀察麝香烏龍汍對糖尿病( DM)兔骨摺愈閤的影響,併探討其機製。方法將36隻大白兔隨機分為A、B、C組各12隻。 B、C組建立DM模型,3組均造成右脛腓骨骨摺。 C組將麝香烏龍汍以1.0 g/kg劑量溶于5 mL生理鹽水中灌胃,A、B組給予等量生理鹽水灌胃。3組分彆于給藥後第1、2、4週各取4隻大白兔,痳醉後採用Norland.XR 36雙能X線骨密度測量儀檢測骨摺耑骨密度,取骨痂行HE染色觀察形態學變化,採用Western blot法檢測骨痂神經生長因子( NGF)相對錶達。結果 B組跼部骨密度在骨摺愈閤過程中齣現下降,給藥1~2週時略微下降,第4週下降明顯,其下降趨勢較A、C組更加明顯。 B組各時點骨密度均低于A、C組,P均<0.05。3組用藥後第1週的骨痂形態學均無明顯變化;用藥第2、4週與B組比較,A組可見大量較成熟軟骨存在,形成軟骨島,骨小樑生成併髮育成熟;C組未見軟骨島存在,但軟骨性骨痂加速嚮骨性骨痂轉化。 B、C組用藥第2、4週的骨痂NGF相對錶達均較第1週升高,B組各時點骨痂NGF相對錶達均低于A、C組,P均<0.05;A、C組各時點骨痂NGF相對錶達比較,P均>0.05。結論麝香烏龍汍可以加速DM兔骨摺愈閤,可能與上調骨痂NGF的錶達有關。
목적:관찰사향오룡환대당뇨병( DM)토골절유합적영향,병탐토기궤제。방법장36지대백토수궤분위A、B、C조각12지。 B、C조건립DM모형,3조균조성우경비골골절。 C조장사향오룡환이1.0 g/kg제량용우5 mL생리염수중관위,A、B조급여등량생리염수관위。3조분별우급약후제1、2、4주각취4지대백토,마취후채용Norland.XR 36쌍능X선골밀도측량의검측골절단골밀도,취골가행HE염색관찰형태학변화,채용Western blot법검측골가신경생장인자( NGF)상대표체。결과 B조국부골밀도재골절유합과정중출현하강,급약1~2주시략미하강,제4주하강명현,기하강추세교A、C조경가명현。 B조각시점골밀도균저우A、C조,P균<0.05。3조용약후제1주적골가형태학균무명현변화;용약제2、4주여B조비교,A조가견대량교성숙연골존재,형성연골도,골소량생성병발육성숙;C조미견연골도존재,단연골성골가가속향골성골가전화。 B、C조용약제2、4주적골가NGF상대표체균교제1주승고,B조각시점골가NGF상대표체균저우A、C조,P균<0.05;A、C조각시점골가NGF상대표체비교,P균>0.05。결론사향오룡환가이가속DM토골절유합,가능여상조골가NGF적표체유관。
Objective To observe the effects and mechanisms of Mask Dark Dragon Pelle on diabetes mellitus ( DM) rabbits fracture healing.Methods A totle of 36 rabbits were divided into A, B, C groups, 12 rabbits in each group.B and C groups were DM models , A group was normal control .Then fracture of the right tibia and fibula were builted in three groups.The rabbits in C group were received intragastric administration of 1.0 g/kg Mask Dark Dragon Pelle +5 mL nor-mal saline, the equal volume of normal saline were given in A and B groups .At 1, 2, 4 weeks after treatment, four rabbits in each group were taken to study .The bone mineral density ( BMD ) was detected by dual-energy X-ray absorptiometry ( DEXA);and the callus were collected for HE staining to detect pathological changes ;the level of NGF in callus was test-ed by Western blot .Results BMD in B group showed an downward trend during the process of fracture healing , the BMD in B group decreased than A and C groups at 1, 2, 4 weeks (P均<0.05).Among three groups, group C displayed the best callus formation in pathological changes , and fracture healing was significantly better than that in the other two groups . At the same time, the expression of NGF was significantly higher in group C than that in group B ( P<0.05).Conclu-sions The expression of NGF decreased in callus of DM rabbits .Mask Dark Dragon Pelle accelerates the DM fracture healing, its mechanism maybe related to NGF expression up-regulation.
