目的:观察电针(EA)对脑缺血再灌注模型大鼠脑组织中海马区F-actin、MAP-2表达的影响,探讨电针治疗缺血性脑卒中的作用机制。方法:选用54只雄性SD大鼠,随机分为假手术组、模型组、电针组各18只;模型组和电针组采用改良Longa线栓法制作缺血再灌注大脑中动脉闭塞(MCAO)大鼠模型,电针组于脑缺血2小时再灌注后开始电针督脉“百会”、“神庭”穴;TTC染色观察动物脑组织梗塞情况及免疫印迹法检测大鼠左侧海马区F-actin、MAP-2蛋白表达情况。结果:①与模型组相比,电针组F-actin、MAP-2蛋白表达均增高(P<0.05);②TTC染色显示,电针组大鼠梗塞面积显著小于模型组(P<0.05),说明经过电针治疗后,其脑组织病理改变减轻,脑部损伤有一定恢复。结论:电针干预后能显著增加大鼠海马区F-actin、MAP-2的表达,有利于脑缺血后突触重塑和神经细胞轴突的再生,从而促进脑卒中后神经功能恢复、改善认知功能。
目的:觀察電針(EA)對腦缺血再灌註模型大鼠腦組織中海馬區F-actin、MAP-2錶達的影響,探討電針治療缺血性腦卒中的作用機製。方法:選用54隻雄性SD大鼠,隨機分為假手術組、模型組、電針組各18隻;模型組和電針組採用改良Longa線栓法製作缺血再灌註大腦中動脈閉塞(MCAO)大鼠模型,電針組于腦缺血2小時再灌註後開始電針督脈“百會”、“神庭”穴;TTC染色觀察動物腦組織梗塞情況及免疫印跡法檢測大鼠左側海馬區F-actin、MAP-2蛋白錶達情況。結果:①與模型組相比,電針組F-actin、MAP-2蛋白錶達均增高(P<0.05);②TTC染色顯示,電針組大鼠梗塞麵積顯著小于模型組(P<0.05),說明經過電針治療後,其腦組織病理改變減輕,腦部損傷有一定恢複。結論:電針榦預後能顯著增加大鼠海馬區F-actin、MAP-2的錶達,有利于腦缺血後突觸重塑和神經細胞軸突的再生,從而促進腦卒中後神經功能恢複、改善認知功能。
목적:관찰전침(EA)대뇌결혈재관주모형대서뇌조직중해마구F-actin、MAP-2표체적영향,탐토전침치료결혈성뇌졸중적작용궤제。방법:선용54지웅성SD대서,수궤분위가수술조、모형조、전침조각18지;모형조화전침조채용개량Longa선전법제작결혈재관주대뇌중동맥폐새(MCAO)대서모형,전침조우뇌결혈2소시재관주후개시전침독맥“백회”、“신정”혈;TTC염색관찰동물뇌조직경새정황급면역인적법검측대서좌측해마구F-actin、MAP-2단백표체정황。결과:①여모형조상비,전침조F-actin、MAP-2단백표체균증고(P<0.05);②TTC염색현시,전침조대서경새면적현저소우모형조(P<0.05),설명경과전침치료후,기뇌조직병리개변감경,뇌부손상유일정회복。결론:전침간예후능현저증가대서해마구F-actin、MAP-2적표체,유리우뇌결혈후돌촉중소화신경세포축돌적재생,종이촉진뇌졸중후신경공능회복、개선인지공능。
Objective:To observe the influence of electro-acupuncture on the expression of F-actin and MAP-2 in hippocampus of rat models with ce-rebral ischemia-reperfusion, and investigate the mechanism of electro-acupuncture treating ischemic stroke. Methods:Chose 54 male SD rats and ran-domly divided into SO group (18 cases), model group (18 cases) and EA group (18 cases), the model group and EA group were molded middle cere-bral artery occlusion (MCAO) by modified Longa suture method, and the EA group began electro-acupuncture therapy on"Baihui"and"Shenting"acupoint of governor vessel in 2 hours after reperfusion;observed the brain infarction of animal by TTC staining and tested the expression of F-actin, MAP-2 in rats' left hippocampus by Western blot. Results:①Compared with the model group, EA group F-actin, MAP-2 protein expression were in-creased (P<0.05);②TTC staining showed that rat infarct size was significantly smaller than the EA group and model group (P<0.05), explained after acupuncture treatment, reduce the pathological changes in brain tissue, brain damage in rats have some recovery. Conclusion:Electro-acupuncture can significantly increase the expression of F-actin, MAP-2 in hippocampus of rats, which is propitious to the remolding of synapse and regeneration of nerve cell axons after cerebral ischemia, so that promotes the recovery of nerve function after stroke and improves cognitive function.
