利用长期大鼠高烟酰胺喂养模型,观察烟酰胺超载对血浆内甲基化反应指标的影响。结果发现,高烟酰胺摄入将伴随高甲基供体消耗,并引起血中甜菜碱水平明显降低,直接证明过量烟酰胺会导致甲基库耗竭。这表明过量的烟酰胺可能通过甲基耗竭诱导氧化性线粒体和 DNA 损伤及 DNA 甲基化样式的改变。
利用長期大鼠高煙酰胺餵養模型,觀察煙酰胺超載對血漿內甲基化反應指標的影響。結果髮現,高煙酰胺攝入將伴隨高甲基供體消耗,併引起血中甜菜堿水平明顯降低,直接證明過量煙酰胺會導緻甲基庫耗竭。這錶明過量的煙酰胺可能通過甲基耗竭誘導氧化性線粒體和 DNA 損傷及 DNA 甲基化樣式的改變。

Long-term high nicotinamide feeding test was performed to investigate the effect of excess nicotinamide on plasma methylation reaction levels. The results shown high nicotinamide intake may lead to a depletion of labile methyl groups. Nicotinamide overload significantly lower plasma betaine levels which suggested the deletion of methyl groups. Therefore, the present study demonstrated that excess nicotinamide may trigger oxidative damage and abnormal DNA methylation patterns due to promote the generation of ROS and its depletion of the methyl donor pool.
