目的:研究乳酸菌发酵液对人结肠癌细胞株LS174T增殖及凋亡作用的影响,为结肠癌的防治提供依据。方法培养嗜热链球菌、保加利亚乳杆菌并制备发酵液,采用四氮唑蓝(MTT)比色法检测其对结肠癌细胞株LS174T增殖活性的影响,设PBS对照;运用AnnexinV-FITC/PI双染法、流式细胞术检测LS174T细胞凋亡率。结果两种发酵液与LS174T细胞株共培养后,与对照组相比,同时点细胞的增殖活性均有降低,其中,保加利亚乳杆菌组与对照组比较差异有统计学意义;流式细胞术显示两组细胞的凋亡率增高,保加利亚乳杆菌组与对照组比较差异有统计学意义。结论保加利亚乳杆菌发酵液对结肠癌细胞体外增殖有一定抑制与促进凋亡的作用,有望应用于结肠癌的防治。

목적:연구유산균발효액대인결장암세포주LS174T증식급조망작용적영향,위결장암적방치제공의거。방법배양기열련구균、보가리아유간균병제비발효액,채용사담서람(MTT)비색법검측기대결장암세포주LS174T증식활성적영향,설PBS대조;운용AnnexinV-FITC/PI쌍염법、류식세포술검측LS174T세포조망솔。결과량충발효액여LS174T세포주공배양후,여대조조상비,동시점세포적증식활성균유강저,기중,보가리아유간균조여대조조비교차이유통계학의의;류식세포술현시량조세포적조망솔증고,보가리아유간균조여대조조비교차이유통계학의의。결론보가리아유간균발효액대결장암세포체외증식유일정억제여촉진조망적작용,유망응용우결장암적방치。
Objective To explore the effect of lactobacillus fermentation broth on the apoptosis and proliferation of LS174T cells .Methods Streptococcus thermophilus(St .) ,Lactobacillus bulgaricus (Lb .) were cultivatd and fermented .MTT colori- metric method was used to determine the inhibition of LS174T cells caused by two fermentation broth ,the PBS control group was set ;the rate of apoptosis were observed by Annexin V-FITUPI method and flow cytometry .Results Compared with the control group ,proliferation activity of St .and Lb .groups were all falling ,Lb .groups was significantly lower than that in the con- trol group at the same time .Flow cytometry analysis showed that the rate of apoptosis of St .and Lb .groups were increased and Lb .groups was significantly higher than that in the control group at the same time .Conclusion The fermentation broth of Lb . has certain inhibitory effect on the proliferation and can promote apoptosis of colon cancer cells ,which can be expected to be ap- plied in colon cancer prevention .
