愈伤组织形成是植物组织培养的重要阶段。钙调素含量在愈伤组织形成过程中发生变化,但钙调素是否参与愈伤组织形成仍不清楚。为此,本文分析了一个钙调素基因CAM5的组织表达模式,结果发现,CAM5基因在拟南芥植株的根、茎、叶、花及幼苗中均有表达。此外,鉴定了拟南芥CAM5基因的纯合突变体 cam5,并观察其叶柄外植体在愈伤组织诱导过程中的表型,结果发现,在愈伤诱导培养基中培养20 d,与野生型相比,突变体的叶柄外植体膨大比较明显。进一步分析显示,培养20 d 后,突变体外植体的鲜重比培养前增加了39倍,而野生型仅增加了16倍。这些结果暗示CAM5基因参与了愈伤组织形成。
愈傷組織形成是植物組織培養的重要階段。鈣調素含量在愈傷組織形成過程中髮生變化,但鈣調素是否參與愈傷組織形成仍不清楚。為此,本文分析瞭一箇鈣調素基因CAM5的組織錶達模式,結果髮現,CAM5基因在擬南芥植株的根、莖、葉、花及幼苗中均有錶達。此外,鑒定瞭擬南芥CAM5基因的純閤突變體 cam5,併觀察其葉柄外植體在愈傷組織誘導過程中的錶型,結果髮現,在愈傷誘導培養基中培養20 d,與野生型相比,突變體的葉柄外植體膨大比較明顯。進一步分析顯示,培養20 d 後,突變體外植體的鮮重比培養前增加瞭39倍,而野生型僅增加瞭16倍。這些結果暗示CAM5基因參與瞭愈傷組織形成。

Callus formation is an important stage during plant tissue culture,in which the content of calmodulin changes.Howev-er,whether calmodulins participate in callus formation still remains unknown.Therefore,in the paper,the tissue expression pat-tern of a calmodulin gene of CAM5 was analyzed.The results show that CAM5 gene is expressed in Arabidopsis roots,stems, leaves,inflorescence and seedlings.In addition,the cam5 homozygous mutant of Arabidopsis CAM5 gene is identified and the phenotype of cam5 petiole explants is observed in the course of callus induction.The results indicate that the callus on the cam5 petiole explants is dilated more obviously than those on wild-type explants after twenty-day culture on callus inducing medium. Further analysis indicates that the fresh weight of cam5 explants increases 39 times after twenty-day culture than before culture, while the wild type increased only 16 times.These results suggest that CAM5 gene is involved in callus formation.
