目的:探讨从人宫颈癌细胞系CaSki中分离培养宫颈癌干细胞球样细胞的可行性,并观察其体外生物学特性。方法采用肿瘤干细胞无血清悬浮培养法富集CaSki细胞系球样细胞,通过体外传代培养、添加血清诱导分化等技术鉴定其干性,利用流式细胞技术检测细胞周期,并应用蛋白印记法及细胞免疫荧光等方法从分子水平检测相关因子的表达。结果从CaSki细胞系中分离到的干细胞球样细胞可以在无血清培养基中悬浮生长成细胞球,并可在体外稳定传代,具有自我更新能力和分化潜能;肿瘤干细胞球样细胞多处于G0/G1期。与其他分子标志物相比,干细胞球样细胞的CD44明显高表达;与亲本细胞相比,干细胞球样细胞的干性相关因子SOX2、OCT4、β-catenin表达较高。结论利用肿瘤干细胞无血清悬浮培养法可以从CaSki细胞系中分离富集到具有稳定肿瘤干细胞性质的细胞群。
目的:探討從人宮頸癌細胞繫CaSki中分離培養宮頸癌榦細胞毬樣細胞的可行性,併觀察其體外生物學特性。方法採用腫瘤榦細胞無血清懸浮培養法富集CaSki細胞繫毬樣細胞,通過體外傳代培養、添加血清誘導分化等技術鑒定其榦性,利用流式細胞技術檢測細胞週期,併應用蛋白印記法及細胞免疫熒光等方法從分子水平檢測相關因子的錶達。結果從CaSki細胞繫中分離到的榦細胞毬樣細胞可以在無血清培養基中懸浮生長成細胞毬,併可在體外穩定傳代,具有自我更新能力和分化潛能;腫瘤榦細胞毬樣細胞多處于G0/G1期。與其他分子標誌物相比,榦細胞毬樣細胞的CD44明顯高錶達;與親本細胞相比,榦細胞毬樣細胞的榦性相關因子SOX2、OCT4、β-catenin錶達較高。結論利用腫瘤榦細胞無血清懸浮培養法可以從CaSki細胞繫中分離富集到具有穩定腫瘤榦細胞性質的細胞群。
목적:탐토종인궁경암세포계CaSki중분리배양궁경암간세포구양세포적가행성,병관찰기체외생물학특성。방법채용종류간세포무혈청현부배양법부집CaSki세포계구양세포,통과체외전대배양、첨가혈청유도분화등기술감정기간성,이용류식세포기술검측세포주기,병응용단백인기법급세포면역형광등방법종분자수평검측상관인자적표체。결과종CaSki세포계중분리도적간세포구양세포가이재무혈청배양기중현부생장성세포구,병가재체외은정전대,구유자아경신능력화분화잠능;종류간세포구양세포다처우G0/G1기。여기타분자표지물상비,간세포구양세포적CD44명현고표체;여친본세포상비,간세포구양세포적간성상관인자SOX2、OCT4、β-catenin표체교고。결론이용종류간세포무혈청현부배양법가이종CaSki세포계중분리부집도구유은정종류간세포성질적세포군。
Objective To isolate and identify sphere-forming stem-like cells from cervical cancer cell line (CaS-ki), and observe its biological characteristics in vitro. Method We enriched sphere-forming stem-like cells using sphere suspension culture system, and confirmed its stem cell-associated characteristics by evaluating the renewal and serum-induced differentiation abilities. Western Blot technology was applied to detect the expression of stem cell-related factors at the protein level. Result The sphere-forming stem-like cells isolated from CaSki can grow into spheres in suspension in serum-free medium, and were able to self-renew. CaSki cancer stem cell-like cells were en-riched in the G0/G1 phase. Compared with other stem cell markers, CD44 has a high expression and can be as the useful marker for cervical cancer cells. Compared with its parental cells, SOX2, OCT4, β -catenin has a high ex-pression of stem cell-related factors. Conclusion We successfully isolated and enriched sphere-forming stem-like cells from cervical cancer cell line CaSki, using serum-free sphere suspension culture system.
