目的:观察半枝莲总黄酮对顺铂化疗经肺癌A-549细胞后肢皮下荷瘤昆明小鼠肾脏细胞凋亡的保护作用,并从抗氧化应激角度初步探讨其中机制。方法索氏法制备半枝莲总黄酮水提物。雄性昆明小鼠50只,随机分为对照组(C组)、荷瘤组(T组)和半枝莲总黄酮低(L组)、中(M组)、高(H组)剂量组,每组10只。T、L、M和H组在右后肢皮下荷瘤两周后,T、L、M和H组以2 mg/kg剂量腹腔注射顺铂,同时L、M和H组分别以100 mg/kg、200 mg/kg、400 mg/kg剂量的半枝莲总黄酮灌胃,连用7 d。完成后处死小鼠,取一侧肾脏检测相关氧化指标,另一侧肾脏固定后石蜡切片免疫组化法观察凋亡相关蛋白Bax及Bcl-2表达。结果 T组MDA含量明显高于C组(P<0.05),L、M、H组MDA含量较T组明显下降(P<0.05);T组中SOD活性及T-AOC较C组明显下降(P<0.05),半枝莲黄酮干预后较T组有所升高(P<0.05);T组肾组织Bax表达较C组明显增强(P<0.05),Bcl-2表达减弱(P<0.05),半枝莲黄酮干预后,各组Bax表达较T组有减弱趋势,Bcl-2表达有增强表现。结论半枝莲总黄酮能够减轻顺铂化疗荷瘤小鼠的肾脏细胞凋亡,这种保护可能是通过降低肾脏氧化应激水平得以实现的。

목적:관찰반지련총황동대순박화료경폐암A-549세포후지피하하류곤명소서신장세포조망적보호작용,병종항양화응격각도초보탐토기중궤제。방법색씨법제비반지련총황동수제물。웅성곤명소서50지,수궤분위대조조(C조)、하류조(T조)화반지련총황동저(L조)、중(M조)、고(H조)제량조,매조10지。T、L、M화H조재우후지피하하류량주후,T、L、M화H조이2 mg/kg제량복강주사순박,동시L、M화H조분별이100 mg/kg、200 mg/kg、400 mg/kg제량적반지련총황동관위,련용7 d。완성후처사소서,취일측신장검측상관양화지표,령일측신장고정후석사절편면역조화법관찰조망상관단백Bax급Bcl-2표체。결과 T조MDA함량명현고우C조(P<0.05),L、M、H조MDA함량교T조명현하강(P<0.05);T조중SOD활성급T-AOC교C조명현하강(P<0.05),반지련황동간예후교T조유소승고(P<0.05);T조신조직Bax표체교C조명현증강(P<0.05),Bcl-2표체감약(P<0.05),반지련황동간예후,각조Bax표체교T조유감약추세,Bcl-2표체유증강표현。결론반지련총황동능구감경순박화료하류소서적신장세포조망,저충보호가능시통과강저신장양화응격수평득이실현적。
Objective To observe the protective effects of total flavonoids from scutellaria barbata on renal injury caused by cisplatin in tumor bearing mice ,and try to investigate its mechanism from the oxidative stress .Methods Total flavonoids was extracted from scutellaria barbata .50 male Kunming mice were randomly divided into control group ( group C ) , tumor group ( group T ) and total flavonoids of Scutellaria barbata low ( group L ) , middle ( M group),high dose group (H group),10 mice in each group.After T,L,M and H group were injected tumor cells for two weeks,T,L,M and H group were injected with cisplatin at the dose of 2 mg/kg,L,M and H group were filled with total flavonoids at the dose of 100 mg/kg,200 mg/kg,400 mg/kg for 7 days,C group and T group were filled with the same volume of normal saline .Then the mice were sacrificed ,the indexes about oxidation in kidney were measured , the expression of Bax and Bcl-2 were observed .Results The content of MDA in group T was increased than that in group C (P<0.05),decreased in group L,M and H than that in group T (P<0.05).The activity of SOD and T-AOC in group T was decreased significantly than in group C ( P<0.05) ,and that was increased in group L ,M and H by flavonoids .The expression of Bax in group T was significantly increased compared with group C ,and the expression of Bcl-2 was decreased .After the intervention of flavonoids , expression of Bax was decreased in L , M and H group compared with group T ,expression of Bcl-2 showed an increasing trend .Conclusion otal flavonoids from scutellaria barbata could reduce renal injury caused by cisplatin in tumor bearing mice ,which should be achieved by reducing re-nal oxidative stress .
