采用2种方法研究了复方糖苷抗新城疫病毒( Newcastle disease Virus, NDV)感染作用。一是鸡胚成纤维细胞( Chicken embryo fibroblast,CEF)培养法,通过观察 NDV CPE的形成及测定 CEF OD值的变化,计算病毒抑制率等指标,综合评价复方糖苷在细胞水平上抗NDV作用,并从直接灭活病毒、阻断病毒吸附与生物合成及释放等环节,探讨了复方糖苷的抗 NDV 途径。二是将复方糖苷稀释成1200,1000,800,600,300μg/mL 5种浓度与100 EID50 NDV液混合后接种9日龄SPF鸡胚,于72 h后统计鸡胚的存活数并测定尿囊液血凝效价( HA),依此判定复方糖苷在鸡胚内抗NDV作用。结果表明:随着药物浓度的增加CPE的特征逐渐减弱,OD值明显升高,病毒抑制率增大,当复方糖苷质量浓度为600μg/mL 时对NDV具有显著的直接杀灭与阻断吸附作用。 HA在1200,1000μg/mL时均为0。表明复方糖苷具有显著的抗NDV感染作用。

채용2충방법연구료복방당감항신성역병독( Newcastle disease Virus, NDV)감염작용。일시계배성섬유세포( Chicken embryo fibroblast,CEF)배양법,통과관찰 NDV CPE적형성급측정 CEF OD치적변화,계산병독억제솔등지표,종합평개복방당감재세포수평상항NDV작용,병종직접멸활병독、조단병독흡부여생물합성급석방등배절,탐토료복방당감적항 NDV 도경。이시장복방당감희석성1200,1000,800,600,300μg/mL 5충농도여100 EID50 NDV액혼합후접충9일령SPF계배,우72 h후통계계배적존활수병측정뇨낭액혈응효개( HA),의차판정복방당감재계배내항NDV작용。결과표명:수착약물농도적증가CPE적특정축점감약,OD치명현승고,병독억제솔증대,당복방당감질량농도위600μg/mL 시대NDV구유현저적직접살멸여조단흡부작용。 HA재1200,1000μg/mL시균위0。표명복방당감구유현저적항NDV감염작용。
Two methods were used to study the inhibition of compound glycosideon replication of Newcastle Disease Virus in cell and embryo. The first was CEF cell cultural method, and through observation of NDV CPE formation and determination of CEF OD value changes, virus inhibition rate was calculated to comprehensively evaluate NDV inhibition of compound glycoside at the cellular lev?el. From direct inactivated virus, blocking virus adsorption and biosynthesis and release of NDV re?sistant approach of compound glycoside were discussed. The second: compound glycoside was dilu?ted into five different concentrations, 1 200, 1 000, 800, 600 and 300μg/mL, and, vaccinated to 9?day SPF chicken embryos after being mixed with 100 EID50 NDV. After 72 h, survival of chicken embryos was added up and HA titer of allantoic fluid was determined to evaluate anti?NDV effect of compound glycoside in chicken embryos. Experimental results showed that with the increase of drug concentration, characteristics of CPE weakened gradually, OD value increased significantly and vi?rus inhibition rate increased, and when concentration of compound glycoside was 600μg/mL , NDV had significant direct killing and block adsorption funtions. In 1 200 and 1 000μg/mL, HA was 0,indcating that compound glycoside had significant anti?NDV effect.
