目的:探讨Syndecan-1基因和HPA-1基因检测在结肠癌侵袭转移、预后评估中的临床价值。方法选择73例结肠癌手术切除的标本,包括结肠癌组织、癌旁组织和正常组织;采用荧光定量逆转录聚合酶链反应检测三组Syn-decan-1mRNA和HPA-1mRNA表达水平。结果结肠癌组织中Syndecan-1 mRNA表达水平显著低于癌旁组织和正常组织(P<0.05),HPA-1 mRNA表达水平显著高于癌旁组织和正常组织(P<0.05);其中癌旁组织Syndecan-1 mRNA表达水平低于正常组织,HPA-1 mRNA表达水平高于正常组织,组间差异有统计学意义(P<0.05)。肿瘤体积、分化程度、浸润程度、淋巴结转移、血行转移、肿瘤TMN分期与Syndecan-1mRNA表达正相关,与HPA-1mRNA表达呈负相关( P<0.05)。 Pearson相关性分析显示,结肠癌组织中HPA-1mRNA表达与Syndecan-1mRNA呈负相关关系(γ=-0.724,P=0.000)。结论 Syndecan-1基因和HPA-1基因检测对判断结肠癌侵袭转移、评价预后有着重要的作用,值得临床借鉴。

목적:탐토Syndecan-1기인화HPA-1기인검측재결장암침습전이、예후평고중적림상개치。방법선택73례결장암수술절제적표본,포괄결장암조직、암방조직화정상조직;채용형광정량역전록취합매련반응검측삼조Syn-decan-1mRNA화HPA-1mRNA표체수평。결과결장암조직중Syndecan-1 mRNA표체수평현저저우암방조직화정상조직(P<0.05),HPA-1 mRNA표체수평현저고우암방조직화정상조직(P<0.05);기중암방조직Syndecan-1 mRNA표체수평저우정상조직,HPA-1 mRNA표체수평고우정상조직,조간차이유통계학의의(P<0.05)。종류체적、분화정도、침윤정도、림파결전이、혈행전이、종류TMN분기여Syndecan-1mRNA표체정상관,여HPA-1mRNA표체정부상관( P<0.05)。 Pearson상관성분석현시,결장암조직중HPA-1mRNA표체여Syndecan-1mRNA정부상관관계(γ=-0.724,P=0.000)。결론 Syndecan-1기인화HPA-1기인검측대판단결장암침습전이、평개예후유착중요적작용,치득림상차감。
Objective To explore the clinical value of Syndecan-1 gene and HPA-1 gene in invasion and metastasis and prognosis of colon cancer .Methods 73 cases of colorectal cancer samples including colon cancer tissues ,paracancerous tissues and normal tissues were selected .Syndecan-1mRNA and HPA-1mRNA expression of the 3 groups were detected by real-time fluo-rescence quantitative RT-PCR.Results The expression level of Syndecan-1 mRNA in colon cancer tissues was significantly lower than that of the paracancerous tissues and normal tissues (P<0.05),the expression level of HPA-1 mRNA in colon cancer tis-sues was significantly higher than that the paracancerous tissues and normal tissues ( P<0.05);the expression level of Syndecan-1 mRNA in paracancerous tissues was lower than that of normal tissues ,HPA-1 mRNA expression level in paracancerous tissues was higher than that of the normal tissues ,the difference was statistically significant (P<0.05).Tumor size,degree of differentia-tion,infiltration, lymph node metastasis , hematogenous metastasis , and TMN staging were positively correlated with Syndecan-1mRNA expression,and were negatively correlated with HPA-1mRNA expression (P<0.05).Pearson correlation analysis showed a negative correlation between the expression of HPA-1mRNA and Syndecan-1mRNA in colorectal cancer (γ=-0.724,P=0.000).Conclusion Detection of Syndecan-1 gene and HPA-1 gene has important significance in evaluating invasion and me-tastasis,predicting prognosis of colon cancer ,and it is worthy of the clinical promotion .
